Immunoassay of Steroids.—Oestrone 3-methylphosphonothiolate, electrostatically complexed with methylated bovine serum albumen (BSA), is an effective immunogen for the production of an antiserum for oestrone 3-sulphate. The phosphorus-containing ester apparently mimics the size of the sulphate, but is more stable. Reductive amination, involving the coupling of 5a-androstane-3,17-dione and of 17/8-hydroxy-5a-androstan-3-one by reduction of their Schiff bases with NaBHsCN, resulted in coupling between the lysine residues of BSA and the C-3 position of the steroid, with up to 40 molecules of steroid per BSA molecule. These antigens led to effective antisera for the steroids. 

Methylated bovine serum albumin (mBSA) (from Sigma) Dissolve in water (1 mg/mL) and store at-20°C. 

Generally, all soluble immunogens or protein-hapten conjugates are mixed with, emulsified in or adsorbed onto adjuvants. The most widespread adjuvants are (i) Freund s adjuvants (ii) aluminum salts and bentonite (iii) methylated bovine serum albumin and, (iv) heat-inactivated Bordetella pertussis.

Water-in-oil emulsions such as Freund s complete adjuvant, Freund s incomplete adjuvant. The complete adjuvant contains QdX- aDlQ6. Mycobacterium tuberculosis bacteria whereas the incomplete does not. Both have the basis of an immune modulator, e.g., branched glucose polymers or methylated bovine serum albumin (Pierce s AdjuPrimeTM). 

The modified DNA (0.25-0.5mg) was electrostatically complexed to equal amounts of methylated bovine serum albumin in phosphate buffered saline (PBS), and emulsified with complete Freund s adjuvant. Immunization of rabbits and purification of IgG fractions were performed as described previously (6,7).

A unique pulvinic acid derivative, methyl bovinate (98), which contains an extra carbon atom in the form of the carbonyl group of an additional 5-lactone ring has been isolated by Besl et al. from cultures of Suillus bovinus 99). Its structure followed from the spectroscopic data, particularly the proton coupled C-n.m.r. spectrum.
