Erythromycin A, the major fermentation component of S. erythraea, is a 14-membered ring macrolide that is used by medicinal chemists as the foundation for building semisynthetic derivatives of erythromycin antibiotics. (Macrohdes inhibit bacteria by interfering with microbial protein biosynthesis.) Semisynthetic macrolides are popular with clinicians because they can be administered orally and have relatively low toxicity. They are often used to treat respiratory tract infections, and have been especially effective against conditions such as Legionnaires disease and community-acquired pneumonia. Chander Mohan was born in 1975 at Dhariwal, Punjab, India. He received his B.Pharm. and M.Tech.(Pharm.) in bulk drugs from Guru Nanak Dev University and the National Institute of Pharmaceutical Education and Research (NIPER), Mohali. After that he worked as senior chemist at Dr. Reddy s Research Foundation, Hyderabad. He then joined Professor M. P. Mahajan s research group in 2002 for his doctoral studies. His research is focused on the synthesis and chemical transformation of C-5/C-6-substituted pyrimidinones. His research interests include synthesis of medicinally important molecules, transition metal-induced transformations in organic synthesis, reaction mechanism and total synthesis of natural products. 

The criticisms in the previous paragraphs lead to a question If Hansch analysis is of such questionable value, then why has an entire chapter of this textbook been devoted to the subject Despite the fading utility of classical QSAR methods such as Hansch analysis, the logic behind Hansch analysis is invaluable to medicinal chemistry. Synthetic chemists in the pharmaceutical industry intuitively consider the ideas used to construct Hansch equations. Ideas such as electronics, sterics, and lipophilicity underlie traditional SAR approaches in the laboratory. Critical analysis of activity data and emphasis on seeking holes in R-group selection are also fundamental to successful SAR on a lead. Through the study of Hansch analysis, all these crucial ideas are presented in a rational framework that helps demonstrate their relevance. Just as importantly, Hansch analysis provides the foundation for the next generation of QSAR comparative molecular field analysis.

In the fall of 1981, my wife, Janet Denlinger and I went to Paris to spend a year in Laszlo Robert and Jacqueline Labat-Robert s Biochemistry of Connective Tissue Laboratory at the Faculty of Medicine of the University of Paris. Laszlo and his wife Jacqueline are well-known researchers in the connective tissue field. The objective was for Janet to carry out research in the collaboration with the Roberts and write her doctoral thesis on the metabolism of hyaluronan in the vitreus and joint. She had already completed her course work at Columbia University. Professor Jean Montreuil, a polysaccharide chemist and Head of the Department of Chemistry at the University of Lille, along with Professor Robert, accepted the sponsorship of Janet s doctoral thesis which she presented in May of 1982 in French, earning a summa cum laude mention along with her degree. Her thesis work laid the foundation of our understanding of how hyaluronan is metabolized in the vitreus and joints of various animal species such as horses, rabbits and monkeys. 

Davidson s lectures on chymistry, delivered in Latin, were devoted mainly to the preparation and uses of medicinal chemicals, with an explanation of the processes and apparatus concerned, and accompanied by practical demonstrations.
