Erythromycin A, the major fermentation component of S. erythraea, is a 14-membered ring macrolide that is used by medicinal chemists as the foundation for building semisynthetic derivatives of erythromycin antibiotics. (Macrohdes inhibit bacteria by interfering with microbial protein biosynthesis.) Semisynthetic macrolides are popular with clinicians because they can be administered orally and have relatively low toxicity. Chander Mohan was born in 1975 at Dhariwal, Punjab, India. He received his B.Pharm. and M.Tech.(Pharm.) in bulk drugs from Guru Nanak Dev University and the National Institute of Pharmaceutical Education and Research (NIPER), Mohali. After that he worked as senior chemist at Dr. Reddy s Research Foundation, Hyderabad. He then joined Professor M. P. Mahajan s research group in 2002 for his doctoral studies. Synthetic chemists in the pharmaceutical industry intuitively consider the ideas used to construct Hansch equations. Ideas such as electronics, sterics, and lipophilicity underlie traditional SAR approaches in the laboratory. Critical analysis of activity data and emphasis on seeking holes in R-group selection are also fundamental to successful SAR on a lead. Through the study of Hansch analysis, all these crucial ideas are presented in a rational framework that helps demonstrate their relevance. In the fall of 1981, my wife, Janet Denlinger and I went to Paris to spend a year in Laszlo Robert and Jacqueline Labat-Robert s Biochemistry of Connective Tissue Laboratory at the Faculty of Medicine of the University of Paris.
