Lipoproteins composition

Partial summary of lipoprotein metabolism in humans. I to VII are sites of action of hypolipidemic drugs. I, stimulation of bile acid and/or cholesterol fecal excretion II, stimulation of lipoprotein lipase activity III, inhibition of VLDL production and secretion IV, inhibition of cholesterol biosynthesis V, stimulation of cholesterol secretion into bile fluid VI, stimulation of cholesterol conversion to bile acids VII, increased plasma clearance of LDL due either to increased LDL receptor activity or altered lipoprotein composition. Lipoproteins interact with enzymes (e.g., lecithin cholesterol acyltrans-ferase, or lipases), with lipid transfer proteins, or with receptors on cell surfaces. The composition of a lipoprotein class depends upon the results of these kinds of interactions. Ultracentrifugation itself may result in minor changes in lipoprotein composition (see Sections 6 and 6.1). 

From this emulsion particle model, it was possible to predict the lipoprotein composition, buoyant density, and hydrodynamic properties of the LDL as a function of lipoprotein size, given the partial specific volumes of the lipid, protein, and carbohydrate components. It is our primary purpose here to emphasize some potentially important revisions in lipoprotein procedure, including additions to the technology of lipoprotein analysis. At the same time, we wish to present some recent data on lipoprotein composition determined by improved lipid analytical techniques. Finally, as a preliminary application of these techniques, we have included the evaluation of correlations between the major variables studied, including all measiued portions of the low- and high-density lipoprotein spectra as observed in an adult male and female population.

Multi-variable or cold-ethanol precipitation techniques yielded plasma protein fractions rich in lipoprotein (Oncley et al. 1949). These isolation procedures were extremely important. They demonstrated that physical properties such as solubility could be studied with lipoproteins as well as other proteins. Electrophoretic mobilities and specific refractive increments of lipoproteins were shown to be measurable properties (Armstrong et al. 1947 a and 1947 b). A detailed analysis of lipoprotein composition was undertaken (Oncley et al. 1950).
