Library construction libraries

Tripeptide libraries have shown activity against certain G-protein coupled receptors. For example, screening a library of ca. 221 000 tripeptides (library 3.19) furnished the potent and selective melanocyte-stimulating hormone agonist, H-D-Trp-Arg-Leu-NH2 (IC50 = 620 nM) [62]. A vasopressin-2 receptor antagonist, H-D-Tic-D-Tyr-D-(4-nitro)-Phe-OH (IC50 = 63 nM), was revealed in a second independently prepared tripeptide library construct (library 3.21 15 625 members) [51]. [Pg.114]

Inhibitors for proteases plasmepsin I and II of the malaria parasite Plasmodium falciparum, with a good plasmepsin/human protease cathepsin D selectivity, have been identified via library construction involving rapid microwave-accelerated Suzuki reactions [57]. The phenyl ring of the biphenyl unit in the lead compound M-((lS)-l- [((lS,2S)-3- [(lS)-2-amino-l-(4-phenyl-benzyl)-2-oxoethyl]amino -2-hydroxy-l-phenoxypropyl)amino]carbonyl -2-methylpropyl)pyridine-2-carboxamide has been altered by performing Suzuki reactions on N-((lS)-l- [((lS,2S)-3- [(lS)-2-amino-l-(4-bromobenzyl)-2-oxoethyl]amino -2-hydroxy-l-phenoxypropyl)amino]carbonyl -2-methyl-propyl)pyridine-2-carboxamide (Scheme 37). In particular, a 2-benzofuryl moiety proved to be interesting since a Ki value of 13 nM for plasmepsin I and... [Pg.174]

All three systems are amenable to sequential substitutions, giving opportunities for use as scaffolds and also, particularly for pyrimidines, rapid muticomponent, often one pot , ring constructions are possible. Both these features give great potential for combinatorial chemistry and library construction. [Pg.383]

Several groups have reported the isolation of cDNA clones for myeloperoxidase from cDNA libraries constructed from mRNA purified from HL-60 cells (Morishita et al. 1987 Weil et al. 1987). If such libraries are induced to express the proteins for which the cDNA molecules encode, then they can be screened for particular genes using antibodies. Indeed, this has been the approach used to isolate cDNA clones for myeloperoxidase from HL-60 cells. Full-length clones have been described and sequenced, and their identity confirmed by a variety of means ... [Pg.62]

This review focuses upon NMR screening strategies and discusses how compound libraries can be designed to provide information pertinent to the specific strategy being pursued. The principles and tools of library construction are described and illustrated by the example of the approach used in our laboratory. The technical details of experimental methods will not be discussed here since they are reviewed in other chapters of this volume and elsewhere [1],... [Pg.391]

One other important consideration in constructing a library is checking that all spectra are correct. This can be done by visual inspection of the spectra and outlier detection tools. For various reasons (e.g. an inadequately filled cuvette, large temperature fluctuations), spectra may differ from others due to factors other than natural variability. Any such spectra should be excluded from the library. Spectral libraries are... [Pg.470]

As mentioned above, in order to avoid creating severely biased libraries, components of comparable reactivity should be used in the library construction. Due to slow exchange rates or unfavorable equilibria, some thiols and acyl components were excluded from further studies. However,... [Pg.179]

Afifymax s thiolacyl library (Fig. 1.9) and Pharmacopeia s statine library (Fig. 1.10) are pharmacophore-based libraries however, their design is different. In the former library, a pool of advanced library intermediates are derivatized with the pharmacophore (thiolacylation) as the final step in library construction, while in the latter library the pharmacophore (statine) is derivatized with synthons as part of library construction. [Pg.15]

Unclick empty linker option to allow addition and modification of the linkers (Fig. 18.6). In this exercise, we only demonstrate enumeration using two linkers. More linkers could be included for chemical library construction. [Pg.353]

The problem of ligand library construction is closely related to the number of synthetic steps required to synthesize a bidentate ligand. These steps are in many cases non-trivial synthetic operations, which renders the ligand synthesis unsuited for automation. A particular challenge is the synthesis of nonsymmetric bidentate ligands equipped with two different donor sites [6,7]. [Pg.30]
