Crude chloroform-methanol-water (30 60 8, v/v) extracts of immunostainedTLC bands were analyzed without further purification by nanoelectrospray low-energy mass spectrometry. Retention of native carbohydrate structure also is important in applications that utilize the conjugated polysaccharide in binding studies with receptors or lectins. A third area of development in carbohydrate l.c. analyses is in the combined techniques (see Section IV,3) and other methods that provide qualitative, as well as quantitative, information about sample constituents, such as high-performance liquid affinity chromatography. The use of specific lectin- and monoclonal antibody-based, stationary phases for analytical and preparative applications is now being considered. The basic concepts of these techniques have been reviewed - and their applications to carbohydrates have been discussed. 

Another interesting and recent application of ACE is the fluorescence-enhanced competition assay for the detection of sugar-lectin interactions (77). Because glucose competes with the fluorescently labeled dextran for the lectin-binding sites, the relative fluorescent intensity due to the displaced dextran is proportional to the concentration of glucose.

The main viral Ag proteins are gp51 and p24. The vaccine from the Leiconad company contains inner proteins, in particular, p24. To differentiate the sick from vaccinated cows the glycolated protein gp51 can be used. It can be immobilized on the transducer surface through the intermediate layer created from various lectins. It was found that among all the lectins tested (PLA, STA, HPA and WGA), PLA and WGA were the most suitable lectins, especially the latter. The application of WGA generates the maximal response of the immune biosensor.
