Application of a computer program to these data of Fig. 24 yielded calculated values of a for every possible solute pair in the seven mobile phases used. The program then interpolated these data over the entire compositional triangle by fitting to a quadratic curve, to yield values of a as a function of mobile-phase composition. Finally, these a values are plotted in trilinear form (Fig. 25) in such a manner as to indicate mobile-phase compositions of optimum selectivity. One of the problems with the diagnosis and treatment of EPM is that the sensitivity and specificity of the tests available currently to diagnose this disease continue to be debated and antibodies to S. neurona may persist in the cerebrospinal fluid (CSF), making it difficult to evaluate treatment efficacy. A negative result on Western blot of a CSF sample is evidence of absence of infection (Daft et al 2002). A new photoconversion method for TEM immunolabeling promises, at least in certain cases, to reduce these problems of penetration and local accessibility. This method is based on the generation of free radicals by excitation of the fluorochrome the oxidation of diaminobenzidine (DAB) by these free radicals generates precipitates that are converted to an electron-dense stain by osmium tetroxide (Deerinck et al., 1994). The ultimate resolution of this method should be determined by the diffusion of the free radicals prior to reaction with DAB and the size of the precipitate initial results indicate resolution comparable to immunogold methods (Deerinck et ai, 1994 Huang et al., 1994), although a detailed comparison of the sensitivity of the photoconversion method has not yet been done. [Pg.108]

The quaternary carbon (ca. ppm) is likewise split, but more narrowly. The methoxyl carbon, like the methoxyl proton, is insensitive to configuration. The 3 Carbons (52-56 ppm) show tetrad and hexad splitting. Most sensitive is the carbonyl spectrum at ca. ITT Ppm, shown expanded in (b) this shows splitting to the base line for most of the pentad sequences, and clear indications of heptad splitting as well the latter observation indicates resolution of the resonances of structures differing only in the relative orientation of carbonyl groups separated by seven intervening carbon-carbon bonds. [Pg.39]

In general, the performance of a radioscopic system should always be checked via test pieces with natural flaws. To ensure an additional comparability, standardized image quality indicators have to be used to control the essential image quality parameters such as spatial resolution (unsharpness) and contrast sensitivity. [Pg.438]

Before the data can be visualised, ie displayed in a two or three-dimensional representation, the ultrasonic responses from the interior of the test-piece must be reconstructed from the raw ultrasonic data. The reconstruction process projects ultrasonic indications into 3D space. As well as reconstructing the entire ultrasonic data set within an acquisition file, it is possible to define an arbitrary sub-volume of the test object over which reconstruction will take place. The image resolution may also be defined by the user. The factor A has been measured for a variety of samples, indicating that the approximation can be applied up to quasi-atomic resolution. In the case of biological specimens typical values of are of the order of 5-7%, as detemiined from images with a resolution of better than 10 A [37,38]- For an easy interpretation of image contrast and a retrieval of the object infomiation from the contrast, such a combination of phase and amplitude hifomiation is necessary. [Pg.1638]

Microscopes are also used as analytical tools for strain analysis in materials science, detenuination of refractive indices and for monitoring biological processes in vivo on a microscopic scale etc. In this case resolution is not necessarily the only important issue rather it is the sensitivity allowing the physical quantity under investigation to be accurately detennined. [Pg.1655]

Any improvement in resolution obtained by increasing ki generally comes at the expense of a longer analysis time. This is also indicated in Figure 12.11, which shows the relative change in retention time as a function of the new capacity factor. Note that a minimum in the retention time curve occurs when b is equal to 2, and that retention time increases in either direction.
