In a study of Finnish workers (292 men and 557 women) exposed primarily to tetrachloroethylene, nonsignificant increases in non-Hodgkin s lymphoma, cervical cancer, and pancreatic cancer were noted (Anttila et al. 1995). The total number of cancer cases in this study was 31. Blood concentrations of tetrachloroethylene measured in random samples taken from these workers averaged 116 pg/L in men and 66 pg/L in women. As the biological exposure index associated with an 8-hour exposure of 25 ppm is 500 pg/L tetrachloroethylene in blood (ACGIH 1995), these workers were probably exposed to concentrations of tetrachloroethylene below 25 ppm. [Pg.67]

Uracil is used more effectively, in nucleic acid synthesis within a rat hepatoma than in normal liver. This observation appears to have stimulated the synthesis of 5-fluorouracil (1027) as an antimetabolite mainly because the introduction of a fluorine atom involves a minimal increase in size. In the event, 5-fluorouracil did prove to have antineoplastic activity and it is now a valuable drug for treatment of tumors of the breast, colon or rectum, and to a lesser extent, gastric, hepatic, pancreatic, uterine, ovarian and bladder carcinomas. As with other drugs which interfere with DNA synthesis, the therapeutic index is quite low and great care is required during treatment (69MI21301). [Pg.152]

In a randomized, multicenter study in 94 patients, mesalazine 4 g/day for 12 weeks in a microgranular formulation was as effective as a standard dose of a glucocorticoid (6-methylpredisolone 40 mg/day) in mild to moderate Crohn s ileitis (Crohn s Disease Activity Index 180-350) (9). The group treated with methylpredisolone had a higher number of adverse events than those given mesalazine. The only adverse effect related to mesalazine was acute pancreatitis, which resolved on withdrawal. [Pg.138]

Test results are affected by defective intestinal absorption caused by intestinal disease or variations in transit time, by impaired hepatic conjugation caused by liver disease, and by impaired renal excretion. To compensate for these possible errors, a control substance (e.g., PABA) may be given on a second day, or alternatively, C-PABA or p-aminosalicylic acid (PAS) can be given orally with the NBT-PABA. Low recovery of the control substance in the urine indicates probable decreased intestinal absorption or decreased renal excretion. Mortality data are frequently used in cohort studies as opposed to incidence data because of the relative ease of obtaining information on deaths. In particular, the advent of the National Death Index has made mortality data more readily available. While all states maintain registries of incident cancer cases, many of the registries are relatively new and data quality can vary from state to state. Investigators conducting follow-up studies are required to comply with each state s requirements for use of the data. For cancers such as pancreatic cancer where survival is poor, mortality data is an excellent surrogate for the risk of the disease. For other cancers where the survival is much better, such as testicular cancer, mortality is a poor estimator of incidence. Table 15.5 describes the 5-year survival for several selected cancer sites for the period 1996-2004. As evident from the table, there is a considerable difference in survival rates for cancer of different sites. The 5-year survival rate for pancreatic cancer was only 5.1% compared to a survival rate of 98.9% for prostate cancer (SEER 2008). [Pg.404]

Inducible NOS has been demonstrated to be overexpressed in chronic pancreatitis and in pancreatic cancer and a significant correlation with COX-2 expression was observed, providing evidence of a link between inflammation and tumor development. Other authors have shown a co-expression of the two inducible enzymes in pancreatic adenocarcinoma (Kong et al. 2002), but the activities of the two enzymes have been found to counteract each other COX-2 overexpression positively correlated with high Ki-67 expression and tumor proliferation, and high iNOS expression was significantly associated with a high apoptotic index. [Pg.71]
