In-process control sampling

Experimental campaign bulk or in-process control samples (e.g., prior to recrystallization) are excellent sources of process-related impurities and are a vital component of the KPSS. Isolated fractions collected from a preparative or semi-preparative liquid chromatography (LC) system are also excellent KPSS samples. Sometimes small-scale synthesis of the impurity or degradant is possible and less time-consuming than the isolation techniques.

The master records should also indicate the sampling and test procedures involved with in-process control and sampling procedures for finished drug products... 

Samples of intermediates and finished products taken for analysis are recorded, stating the time, date, and conditions for these samples. Deviations in operating conditions and out of specification (OOS) conditions in samples are reported and investigated. Figure 9.2 shows a mechanism for production and in-process controls. 

This definition can be described as analysis in the process and is closely related to the traditional role of analytical chemistry in process control. The classical scope of a process analytical method is it to supplement the control scheme of a manufacturing process with data from a process analyzer that directly measures chemical or physical attributes of the sample. 

Neither one-point nor two-point calibrations have room to test the model or statistical assumptions, but as long as the model has been rigorously validated its use in the laboratory has been verified, these methods can work well. Typical use of this calibration is in process control in pharmaceutical companies, where the system is very well known and controlled and there are sufficient quality control samples to ensure on-going performance. 

The in-process inspection and testing is carried out in accordance with the approved test methods, work instmctions, and qualified equipment. For in-process control, statistical sampling techniques are used as appropriate. 

For in-process control and determination of quality levels, inspection plans and quality control charts are used (as relevant). For sampling plans and acceptance criteria. Military Standard 105E is used. 

BULL The other thing I would like to do is reinforce what Cotruvo said about the use of surrogate methods in process control. I think they are probably very useful in a situation where you know what is going on. But when you are trying to take a TOX measurement, which might include anything from dioxin in one sample to trichloroethylene in another, the meaning of that TOX result is entirely different in the two circumstances. 

In-process controls such as stratined sampling, process analytical technology (PAT) application, and blend homogeneity. Assess modincation of dissolution through optimization of API characteristics and then perform assessment of specialized technologies [hot-melt extrusion (HME), spray-dried dispersion, solid dispersion, etc.] for long-term resolution... 

Production and in-process controls (unit operations, time limits, in-process sampling, blending of intermediates or APIs, contamination control)...
