The cells are cultured and in vitro restimulated with antigen (here PT) for 12 h to mimic the encounter with the specific antigen in the body. Earlier studies have shown that an in vitro restimulation of mouse spleen lymphocytes with antigen for 4 and 24 h, can be used to study immune response as well as other cellular processes, such as apoptosis, inflammation, and so on (see Chapter 16). Our experiments, however, showed that immune response markers are expressed at later time-points as compared to mouse, and that 12 h of in vitro restimulation is the ideal time-point for the assessment of immune response genes (see Table 17.1) (see Note 1).

C4 is coded by two nonallelic loci in the major histocompatibility complex region of chromosome 6p, closely linked to the genes for C2, factor B, and HLA-DR. The close linkage to the immune response genes may account for most of the disease associations reported for C4 variants and deficiency, rather than abnormalities of C4 itself.

Class II MHC antigens are found only on certain lymphoid cells. These are products of (mouse) la and Ir (for immune response) genes. 

The I (Immune response) region of the H-2 complex was initially defined by the studies of McDevitt and co-workers on the Ir-1 genes. These immune response genes are reviewed in detail by Benacerraf in Chapter 4 of this volume. Therefore, we will simply summarize their principal features.
