Glucose, determination

We do not use this procedure for glucose, but Figure 37 shows a series of glucose determinations being performed on 25 microliters of blood, illustrating the fact, that with this system, there may be no wash-out from specimen to specimen. 

Measurements in living things, e.g. patients, is occasionally done by sampling in a non-invasive way. Because no standard patients are available which can be selected for calibration, another principle of calibration must be applied, e.g., by a reference method that measures parallel to the actual measuring method. So, the calibration consists in a comparison of the measured results with that of the reference method which are considered to be true (that have been validated beforehand). An example of a reference calibration is represented by non-invasive blood glucose determination by means of NIR spectroscopy (Muller et al. [1997] Fischbacher et al. [1997] Danzer et al. [1998], see Sect. 6.2.6).

Niacin requires baseline tests of liver function (alanine aminotransferase), uric acid, and glucose. Repeat tests are appropriate at doses of 1,000 to 1,500 mg/day. Symptoms of myopathy or diabetes should be investigated and may require creatine kinase or glucose determinations. Patients with diabetes may require more frequent monitoring.

Mix glucose determination reagent concentrate (Total Starch Assay Kit or Glucose Assay Kit Megazyme International Ireland) with 1 liter of lx phosphate buffer (see recipe). Store in aliquots, protected from light, for 3 months at 2° to 5°C or >1 year at -20°C. 

The analysis of maltose syrups is carried out similarly to that of glucose, determinations of the reducing sugars and dextrin being of particular importance. With malt extracts, a determination of the diastatic power may also be required.
