Libraries genomic

Many clones must be created to be confident that the genomic library contains the gene of interest. The probability, P, that some number of clones, N, contains a particular fragment representing a fraction, f, of the genome is... [Pg.406]

Yeast (Saeeharomyees eerevisiae) has a genome size of 1.21 X 10 bp. If a genomic library of yeast DNA was constructed in a bacteriophage A vector capable of carrying 16-kbp inserts, how many indi-... [Pg.422]

Upon screening genomic libraries obtained from environmental samples, more than 200 new nitrilases that allow mild and selective hydrolysis of nitriles were discovered [56]. One of them catalyzes the (R)-selective hydrolysis of (16) with a value... [Pg.40]

Library A collection of cloned fragments that represents the entire genome. Libraries may be either genomic DNA (in which both introns and exons are represented) or cDNA (in which only exons are represented). [Pg.413]

As an attempt to identify the genes of the secondary pectinases, we screened a genomic library of the E. chrysanthemi strain 3937 for pectinolytic activities and identified the gene of a novel PME. We propose to call this gene pemB and to rename pemA the gene coding for the already characterised PME which was previously called pern (3,4). [Pg.837]

At this time, more then thirty channel DNAs have been cloned and characterized from various sources, predominantly from Drosophila melanogaster, mouse, rat and human cDNA/genomic libraries [6-31]. Inspection of the derived primary K channel protein sequences indicates that voltage-gated channels belong to a... [Pg.297]

Genomic library A collection of clones made from a set of randomly generated overlapping DNA fragments representing the entire genome of an organism. [Pg.534]

DeSantis et have reported the discovery of new nitrilases through the screening of genomic libraries created by the extraction of DNA from various environments (metagenomics). In preliminary experiments, using 25 mM mandelonitrile in pH 8 buffer containing 10% methanol and 0.12 g mL of one of these nitrilases, the acid was produced quantitatively with 98 % ee within 10 min. The product was subsequently shown to be (7 )-mandelic acid after isolation in 86 % yield. In a parallel reaction, (/ )-2-chloromande-lic acid was produced at a seventeenth of the rate (Scheme 1.44). [Pg.44]
