Functional selectivity

This discussion will be limited to functions of one variable that can be plotted in 2-space over the interval considered and that constitute the upper boundar y of a well-defined area. The functions selected for illustration are simple and well-behaved, they are smooth, single valued, and have no discontinuities. When discontinuities or singularities do occur (for example the cusp point of the Is hydrogen orbital at the nucleus), we shall integrate up to the singularity but not include it. 

The achiral triene chain of (a//-rrans-)-3-demethyl-famesic ester as well as its (6-cis-)-isoiner cyclize in the presence of acids to give the decalol derivative with four chirai centres whose relative configuration is well defined (P.A. Stadler, 1957 A. Escherunoser, 1959 W.S. Johnson, 1968, 1976). A monocyclic diene is formed as an intermediate (G. Stork, 1955). With more complicated 1,5-polyenes, such as squalene, oily mixtures of various cycliz-ation products are obtained. The 18,19-glycol of squalene 2,3-oxide, however, cyclized in modest yield with picric acid catalysis to give a complex tetracyclic natural product with nine chiral centres. Picric acid acts as a protic acid of medium strength whose conjugated base is non-nucleophilic. Such acids activate oxygen functions selectively (K.B. Sharpless, 1970).

Mice models reveal that both PR foims are physiologically important. Mice lacking the PR gene fail to ovulate, are infertile, and have impaired thymic function. Selective PRA-deficient female mice are infertile due to reduced oocyte and uterine deficiency... 

However, more recently, a functionally selective inhibitor of nNOS has been described — 7-nitroindazole (7-NI). It is puzzling that in vitro this compound has no selectivity for nNOS over eNOS but after systemic administration, fails to change blood pressure yet alters neuronal responses that are thought to result from production of NO. A suggested resolution of this action is that 7-NI is metabolised in the periphery but not the CNS, so that once it has crossed the blood-brain barrier, it can only act on nNOS.

The best studied of the endocarmabinoids are anandamide (A -arachidonyl-ethanolamine, AEA)(1) and 2-arachidonylglycerol (2-AG)(2). Anandamide was first identified from porcine brain extracts by Devane and co-workers in 1992 [13], while 2-AG was first reported in 1995 to have been isolated from canine gut [14] and rat brain [15]. More recently, noladin ether (2-arachidonyl-glyceryl ether, 2-AGE)(3) [16], virodhamine (D-arachidonyl-ethanolamine)(4) [17] and A-arachidonyl-dopamine (NADA)(5) [18] were proposed as endogenous ligands for the cannabinoid receptors. In a subsequent publication, the authors failed to detect noladin ether in mammalian brains and questioned the relevance of this compound as an endocarmabinoid [19]. Anandamide, noladin ether and NADA have functional selectivity for CBi receptors, virodhamine is CB2 selective and 2-AG is essentially non-selective.

Using our dataset which includes all of the descriptors mentioned so far, we conducted a PLS analysis using SIMCA software [34], In the initial PLS model, MW, V, and a (Alpha) were removed because they are in each case highly correlated with CMR (r > 0.95). SIMCA s VIP function selected only qmin (Qnegmin) for removal on the basis of it making no important contribution to the model. In the second model, 2q+/a (SQpos A) and ECa/a (SCa A) coincided nearly exactly in the three-component space of these two, we decided to keep only ECa/a in the third and final model. This model consisted of three components and accounted for 75% of the variance in log SQ the Q2 value was 0.66.

The quinoxalinedione derivatives 6-cyano-7-nitroquinoxaline (CNQX), 6,7-dinitro-quinoxaline-2,3-dione (DNQX), and 2,3-dioxo-6-nitro-l,2,3,4-tetrahydrobenzo[f]quinox-aline-7-sulphonamide (NBQX) are potent competitive antagonists of AMPA and kainate receptors. Of these, NBQX shows the most functional selectivity (approximately threefold) for AMPA over KA receptors (45), and has been used in low concentrations (1 pM) to isolate kainate currents in hippocampal interneurons (46).
