Salmine Sulfate. A protamine found in the sperm of salmon. Contains arginine, proline, serine, glycine, valine, leucine, alanine, threonine, isoleucine, lysine, histidine, aspartic and glutamic acids. Mol wt 6000 to 7000. N about 25%. While sulfur-free amino acids are broken down to amines via decarboxylation, the sulfur-containing amino acids such as cysteine can undergo more complex reactions. Keilin and Mann (1939, 1940a) first noted that zinc is a constituent of carbonic anhydrase. Leiner and Leiner (1940) and Hove, Elvehjem, and Hart (1940) corroborated these findings. Day and Franklin (1946) found zinc in carbonic anhydrase extracted from plants. The figures on the percentage zinc content vary. Keilin and Mann (1940a) found 0.33 %, Hove, Elvehjem, and Hart (1940) 0.318% for enzyme preparations of comparable purity, as judged by their activity. Jensen and DeLawder9s purified commercial insulin by adsorption on activated carbon followed by elution with 90% phenol. The phenol eluate was diluted with water and the insulin that flocked out was washed with absolute alcohol and ether. Insulin thus prepared has an activity about equal to that of crystalline insulin. The activity was not further increased by a subsequent adsorption and elution. This finding is supported by data of Fisher and Scott,96 and is in contrast to an observation by Dingemanse97 that a product of greater activity than crystalline insulin can be prepared by this procedure. [Pg.293]
