Experience full-time work

At that time we became involved full time working on transcarboxylase so I asked Pat Siu, one of my graduate students, to take a hand with this problem. About 3 days later he came to me and said, I don t know what is wrong with you and Stjernholm, I can fractionate the crude extract with ammonium sulfate and the fraction is active with no further additions . I said, Ah, come on, what are you talking about. Rune and I have done those experiments a dozen times and never got activity . But Pat refused to be swayed, so we went over his protocol step by step. Then we discovered that Pat Siu had dissolved the ammonium sulfate fraction in phosphate buffer whereas Stjemholm and I had used Tris buffer. Thus, Siu had shown that phosphate is required for this reaction and then and there we wrote down the reaction ...

In order to conceive the contributions of the various species to every band, the spectra are deconvoluted.
