PGA was one of the very first degradable polymers ever investigated for biomedical use. PGA found favor as a degradable suture, and has been actively used since 1970 [45 -7]. Because PGA is poorly soluble in many common solvents, limited research has been conducted with PGA-based drug delivery devices. Instead, most recent research has focused on short-term tissue engineering scaffolds. PGA is often fabricated into a mesh network and has been used as a scaffold for bone [48-51], cartilage [52-54], tendon [55, 56], and tooth [57]. [Pg.72]

Since 1988, the methods that we use to isolate cDNAs of alkaloid biosynthesis have become ever more facile and sensitive, allowing for more efficient cDNA identification. We do not, however, yet understand enough about the cellular localization of alkaloid formation or about the nature of the catalysts to move completely away from enzymology and biochemistry and to use only molecular genetic techniques to dissect these biosynthetic pathways. Even our most recently successful cDNA isolations and identifications involved classical protein purification. We are beginning now to use proteomics and EST sequencing to identify natural product biosynthetic cDNAs, but these approaches are more feasible when a specialized cell/tissue type in which secondary metabolite biosynthetic pathways are active, can be isolated and used as a protein or RNA source. [Pg.176]

Albert H. Coons was the first to attach a fluorescent dye (fluorescein isocyanate) to an antibody and to use this antibody to localize its respective antigen in a tissue section. Fluorescein, one of the most popular fluorochromes ever designed, has enjoyed extensive application in immunofluorescence labeling. For many years, classical fluorescent probes such as FITC or Texas red (TR) have been successfully utilized in fluorescence microscopy. In recent decades, brighter and more stable fluorochromes have continually been developed (see Table 14.1). Marketed by a number of distributors, cyanine dyes, Cy2, Cy3, Cy5, Cy7, feature enhanced water solubility and photostability as well as a higher fluorescence emission intensity as compared to many of the traditional dyes, such as FITC or TR. [Pg.137]

It is obvious that one should not expect to find in the literature extensive information regarding the composition of the brains, livers, or even muscles of healthy human individualsespecially so since repeated samples would have to be taken for analysis in order to determine conclusively the importance of inter-individual differences. The best that could be hoped for would be extensive "horizontal" studies relative to the composition of blood, secretions, etc., of individual human specimens and, perhaps, more comprehensive data including tissue composition with respect to animals. However, satisfactory studies of this sort have seldom, if ever, been made. More often than not, such horizontal studies as have been made have not been published in complete enough form to give the kind of information needed to answer the questions which we are considering. [Pg.73]
