Each paraffin-embedded section is collected on microscope slides and first examined under a microscope (lOx) to ensure that it contained sufficient tumor material and to eliminate possible contaminating normal tissues. Tumor and tumor-free areas are identified within 15 pm-thick deparaffinized sections lightly counterstained with hematoxylin and microdissected by gentle scraping with sterile scalpels into 1.5 ml polypropylene vials, using a hematoxylin and eosin-stained step section from the same block [4-7].

Cryo-microtomed tissue sections, nominal thickness 5 pm, were mounted on 0.5 mm thickness BaF2 windows and their spectra were recorded in transmission mode. Parallel sections were stained conventionally (with haematoxylin and eosin H E) to facilitate the identification of regions of particular interest.

When an excessive amount of emulsion is administered to the hepatic artery, the excess can be leaked to the vein. The W/O/W and O/W emulsions with and without FARM were administered via the tail veins of male Wistar rats and the acute toxicities of the emulsions were evaluated (Flino et al., 1997). Fifteen days after administration or after they died, the rats were dissected. Macroscopic pathological appearances and optical microscopic changes in hematoxylin-eosin (HE)-stained tissues of lung, stomach, and thymus gland were observed.

For light microscopic examination, liver tissue was fixed in 10 % buffered formalin, embedded in paraffin, sectioned and stained with hematoxylin and eosin. In some cases, preparations were stained with PTAH (phosphotungstic acid-haematoxylin), by the Van Gieson method and the PAS (periodic acid-Schiff)...
