Early Achievements

The idea of isoelectric fractionation of ampholytes has been known for a long time and was described in many articles before a technique became available for practical work. The difficulties which delayed development and application were technical. In particular, it was difficult to achieve a stable pH-gradient and prevent convection in the electrolyte system.

In spite of the slow development of crystal structure analysis, once it did take olT it involved a huge number of investigators tens of thousands of crystal structures were determined, and as experimental and interpretational techniques became more sophisticated, the technique was extended to extremely complex biological molecules. The most notable early achievement was the structure analysis, in 1949, of crystalline penicillin by Dorothy Crowfoot-Hodgkin and Charles Bunn this analysis achieved something that traditional chemical examination had not been able to do. By this time, the crystal structure, and crystal chemistry, of a huge variety of inorganic compounds had been established, and that was most certainly a prerequisite for the creation of modern materials science. 

In light of the fact that this topic was reviewed in 1999 [1], in this chapter I shall simply highlight a few of the particularly noteworthy early achievements, and then describe recent progress, including efforts from our laboratory at MIT.

After the very early achievements due to the real pioneers of chemical topology who forty years ago first acceeded to that intriguing molecule that is a catenane (by statistical threading approach Wasserman, 1960 or by a directed approach Liittringhaus and Schill, 1964), this peculiar field of research concerned with molecules whose prime feature rests without doubt on beauty and non trivial spacial arrangement, has in the beginning of the 80 s literaly exploded with the apparition of various templated approaches.

Gas-phase nucleophilic displacement reactions have been extensively studied both by theory and by experiment. Since an excellent review on gas-phase nucleophilic displacement reactions was published in 1985226, the early achievements in this field are only briefly outlined here to provide some background for the most recent developments.

The development of fast and accurate procedures for the determination of calcium in biological materials represents one of the important early achievements of atomic absorption spectroscopy. The diflBculties encountered with calcium in emission flame photometry are well known (Dll, L6, S6, SIO), but spectral interferences and extreme dependency on flame temperature, serious obstacles in emission, are either nonexistent or of lower importance in absorption. In patients with STE-ACS, early achievement of an open infarct-related artery (the open artery theory) is associated with an improved outcome. Combined use of microbial enzymes as biocatalysts with chemical synthesis has its origin in the steroid transformation developed in the USA in the early 1950s. Arima and his group [11] invented a unique microbial conversion process, in which the aliphatic side-chain of cholesterol was cleaved to produce a steroid core as a starting material for chemical synthesis of steroid hormones. Yamada et al. discovered the reverse reaction of the pyridoxal-containing L-amino acid lyases and applied them to synthesize L-tryptophan and l-DOPA [12] from pyruvate, ammonia and corresponding aromatic compounds. Since these early achievements, a variety of unique processes with newly screened microbial enzymes as biocatalysts have been invented. 

One of the first early achievements of modelling was to assist in the solving of the X-ray structure of DNA. The interpretation of tautomeric forms, the modelling of hydrogen bonds and conformational analysis all had a part to play in suggesting a structure consistent with the true experimental result, the diffraction pattern. As others have pointed out, all atomic structures of proteins are in fact models of experimental data and require modelling tools (force fields) to build them. 

In the Committee discussions, considerable concern was expressed regarding the use of the concept of technical feasibility as the basis of ambient air standards. The Committee determined that 1) the health of people is more important than the question of whether the early achievement of ambient air quality standards protective of health is technically feasible and 2) the growth of pollution load in many areas, even with the application of available technology, would still be deleterious to public health.

Prigogine s teacher De Bonder was the founder of The Brussels School of Thermodynamics that focused on the thermodynamic treatment of irreversible processes, including chemical reactions. Much of the work of The Brussels School was connected with rates of entropy-production of systems that are not at equilibrium. One of Prigogine s early achievements, after he was appointed to the ULB physical chemistry faculty, was to demonstrate that non-equilibrium systems that are close to equilibrium necessarily will evolve so as to approach the equilibrium state in such a way that the rate of entropy production is as low as is possible.
