Doxycycline

Tetracyclines are produced by various Streptomyces strains and are extensively applied in human and veterinary medicine. They display a broad spectrum of antimicrobial activity in combination with low toxicity and can be applied orally. The isolation of the 6-deoxytetracyclines (44) led to other chemical modifications of (1). 6P-Deoxytetracycline [5614-03-9] (13), prepared by catalytic hydrogenolysis of tetracycline (1), resulting ia an iaversion (45) of the configuration at the C-6 position, but retention of antibacterial activity. Catalytic reduction (7,8) of the 6-methylene derivative (14) yields both the 6a-methyl (15) and 6P-methyl compound (13). The 6a-isomer (15) is reported (7,45) to be more active than the 6P isomer (13). The a-isomer, doxycycline (6), is an example of a semisynthetic tetracycline that has become commercially useful. [Pg.179]

In the United States, the manufacturers of fermentation-derived tetracyclines (1), (2), and (3) are the Ledede Laboratories, a division of American Cyanamid Co., Charles Pfizer Inc., Bristol Laboratories, and RacheUe Laboratories. There are also several manufacturers abroad. Tetracycline is now sold genetically by many companies. Pfizer s doxycycline (6) and Ledede s minocycline (7), both semisynthetic tetracyclines, are the only members of the group that have increasing sales. Tetracycline and its derivative doxycycline are antibiotics widely used in the treatment of bacterial infections. They also exert an antimalarial activity. Tetracyclines inhibit the binding of aminoacyl-tRNA to the ribosome during protein synthesis. [Pg.172]

Tetracyclines. It is important to give the tetracyclines on an empty stomach tetracyclines are not to be taken with dairy products (milk or cheese). The exceptions are doxycycline (Vibramycin) and minocycline (Minocin), which may be taken with dairy products or food. The nurse should give clindamycin with food or a full glass of water. The nurse can give troleandomycin and clarithromycin without regard to meals. All tetracyclines should be given with a full glass of water (240 mL). [Pg.88]

Malaria is transmitted from person to person by a certain species of the Anopheles mosquito. The four different protozoans causing malaria are Plasmodium falciparum, P. malariae, P. ovale, and P. vivax. Drugp used to treat or prevent malaria are called anti malarial drags. Three antimalarial drugs are discussed in the chapter chloroquine, doxycycline, and quinine sulfate. Chloroquine (Aralen) is also used in die treatment of extraintestinal amebiasis (see section on Amebicides). Doxycycline is also used to treat infections caused by Neisseria gonorrhoeae, Treponema pallidum, Listeria monocytogenes, Clostridium, and Bacillus anthracis when penicillin is contraindicated. Quinine also may be used for die prevention and treatment of nocturnal leg cramps. [Pg.143]

Doxycycline (Vibramycin) is an antibiotic belonging to die tetracycline group of antibiotics. The adverse reactions associated with this drug are discussed in Chapter 9 and include photosensitivity, anorexia, nausea, and vomiting. [Pg.143]

Doxycycline This drug can cause photosensitivity. Even relatively brief exposure to sunlight may cause sunburn. Avoid exposure to die sun by wearing protective clodiing (eg, long-sleeved shirts and wide-brimmed hats) and by using a sunscreen. [Pg.145]
