Dose-limiting

CI-979 (29) is a balanced muscarinic agonist having equal affinities for cloned ml and m2 receptors (144). However, unlike prototypical muscarinic compounds such as (25), (29) increases central muscarinic tone, as indicated by behavioral and electroencephalogram (EEG) parameters, at doses lower than those requited to produce gastrointestinal effects (144). CI-979 is well tolerated in humans up to a dose of 1 mg. Dose-limiting side effects such as stomach pain and emesis were observed at a dose of 2 mg.

Dactinomycin, an antineoplastic dmg, was discovered in 1943 and is made in rather pure form by StreptomjcesparvuUus. Dactinomycin has some bacteriostatic antibacterial and antifungal activity, but high toxicity limits its use to antineoplastic therapy. It may be used alone or with other antineoplastics, with or without surgery and synergistic x-ray therapy. Dose limiting bone marrow toxicity may result in low white cell and platelet count. Where these limits are impracticable having regard to the nature of the work the employer may apply a dose limit of 100 mSv in any period of 5 consecutive months subject to a maximum effective dose of 50 mSv in any single calendar year, and to prior approval by the Radiation Protection Adviser, the affected employee(s), and the Health and Safety Executive. 

Conduct work in designated controlled areas (e.g. in UK these are areas in which instantaneous dose rates >7.5 pSv/hour occur, or where employees may exceed 6 mSv annual dose limit, or where air concentration or surface contamination exceeds specified levels). 

Investigate accidents which may have led to persons receiving effective doses in excess of 6 mSv or an equivalent dose greater than 30% of any relevant dose limit. Investigate and report to the authorities loss of materials from accidental release to atmosphere, spillages, theft. The Regulations provide a comprehensive list of notifiable concentrations for each radionuclide isotope.

To ensure that during normal operation, maintenance and decommissioning, and in emergency situations, the radiation exposure to workers and the public is kept below the dose limits given in the Basic Safety Standards for Radiation Protection (BSS).
