Diamine sulfate

When p-ethylaminobenzoate and N,N-dimethyl-p-phenylene diamine sulfate were similarly treated with the dibromosulfone (I) for 2 h, the yield was only 32% and 15% respectively (Figure 1). A possible explanation is that the nucleophilicity of these monosubstituted anilines is weaker than that of aniline while a Na2C03 1,4-HBr elimination reaction could be competing with the substitution reaction, leading to the lower yield (18).

PPD is manufactured on a large scale by the US chemical giant Du Pont and is mainly used to manufacture resins and polymers. That which is destined for inclusion in hair dyes is specially made by a different process that ensures it is absolutely pure and that it is free from any by-products that could cause adverse effects. What this means is that if there is a risk to health from exposure to PPD then it will be due to the PPD itself and not to some hidden cause. There is an alternative to PPD and that is TDS (short for toluene-2,5-diamine sulfate). This is less skin sensitive but produces shades of brown that are slightly redder.

To a stirred, ice-cooled suspension of NaCl (1 g) in CISO3H (30 mL) was added in portions quinazolme-2,4-diamine sulfate (13.85 g, 0.05 mol). The resulting mixture was heated at 150-160 °C for 3 h, cooled, and then poured onto ice. The resulting precipitate was collected and washed with H O to afford 2,4-diamino-quinazoline-6-sulfonyl chloride as the sulfate salt yield 19.1 g (99%) mp >300°C,... 

To formamide (20 mL) was added the crystalline sulfate of the appropriate pyrimidine-4,5-diamine (2.0 g). The sulfate salt was prepared in all cases by the addition of dil H2SO4 to an aqueous solution or suspension of the 4,5-diamine. The sulfate precipitated almost immediately in most instances however, in some cases, the salt crystallized slowly only after extended cooling of the aqueous solution. The formamide solution of the pyrimidine-4,5-diamine sulfate was refluxed for 20-25 min. A solution was rapidly obtained and precipitation of the purine occurred after 10 min. The solution was finally cooled, diluted with H O(10 mL) and allowed to remain overnight in the refrigerator, The crude product was removed by filtration from the neutral solution and washed with cold water.
