In comparative studies, [ C]toluene 2,4-diamme, the hydrolysis product of [ ]-toluene 2,4-diisocyanate, was administered to rats at doses of 3 mg/kg bw (orally or intravenously) and 60 mg/kg bw orally. After 48 h, the distribution of radioactivity was similar in all cases (urine, 64-72% faeces, 20-31% and tissue/carcass/gastrointestinal tract, 2-5%). Comparison of the toluene 2,4-diisocyanate inhalation group with the oral toluene 2,4-diisocyanate and toluene 2,4-diamine treatment groups indicated that a larger percentage of the inhaled radioactivity was in the tissues/carcass and that excretion of radioactivity into the urine was slower following toluene 2,4-diisocyanate inhalation. 

Inhalation exposure to toluene 2,4-diisocyanate results primarily in the formation of acid-labile conjugates, with little or no toluene 2,4-diamine being formed. This suggests that the disposition of inhaled toluene 2,4-diisocyanate is quite different from that of orally administered toluene 2,4-diisocyanate or of intravenously or orally administered toluene 2,4-diamine. 

GC-MS can be used for determination of A-trifluoroacetyl derivatives of PAA. In cases when various analytes are present in the sample, MS detection in the scan mode instead of the SIM mode allows further identification by display of the full MS. A study was carried out of water spiked with aniline (la), toluene-2,4-diamine (2e), toluene-2,6-diamine (2f), 4,4/-diaminodiphenylmethane (5a), 4,4/-oxydianiline (6a), 3,3 -dimethylbenzidine (7d), 1,5-diaminonaphthalene and 1,3-phenylenediamine (lk), which underwent SPAD to the corresponding trifluoroacetamides (Section III.B.l). The LOD were in the 0.1 to 0.4 ftgL-1 range, with RSD from 4 to 17%. The method was used for testing the diffusion of PAA into food (simulated by water) which was in contact with laminated flexible food packaging materials. Detectable extracted levels were found for 2e, 2f and 5a68.

When one part of toluene-2 4-diamine is baked with 2 to 4 parts of sulphur at a temperature between 190 and 250 C (374 to 482 F) for 15 to 24 hours, and the crude product is extracted with aqueous caustic soda or sodium sulphide, a sulphur dye known as Immedial Orange C (C.I. SULPHUR ORANGE 1) is extracted. The dye, when heated with an alkali, yields an ortho-aminothiol which can be isolated in a pure state in the form of its lactam after it has been treated with chloracetic acid, and it has been established that it has the structure shown in (2).
