Hodgkin s disease accounts for 1% of all new cancers diagnosed in Western countries and for 15% of all malignant lymphomas. In patients with early stage lA-IIA disease without B-symptoms or bulky adenopathy, therapy consists of either extended field radiotherapy or limited duration chemotherapy, e.g. ABVD (anthracycline, bleomycin, vinblastine, dacarbazine) for 3-4 cycles followed by involved field radiotherapy. Radiation alone results in a 10-year relapse free survival of 70-75% and, because of the efficacy of salvage chemotherapy for those who relapse, an overall survival of 80-85%. The combined modality approach results in fewer relapses but overall survival is similar. In order to reduce the long term morbidity of radiation current trials are exploring combined modality treatment with lower radiation doses versus chemotherapy alone. [Pg.722]

The available preclinical and clinical data strongly support the introduction of antiangiogenic drugs into combined modality treatment regimens that include radiation therapy. [Pg.39]

The available recent preclinical and clinical data strongly support the introduction of antiangiogenics into combined modality treatment schemes that include radiotherapy. The ultimate benefit of these therapeutic combinations needs to be determined with longer follow-up of the effect of these antiangiogenic agents... [Pg.48]

In order to inactivate a tumor cell, several distinct yet overlapping pathways may be activated. Besides the induction of pure apoptosis, other cell inactivation modalities, including programmed necrosis, mitotic catastrophe, senescence, or terminal differentiation, may be triggered (Belka 2006). The influence of a combined modality treatment on any of these end points has never been analyzed in greater detail thus, only very few data are available showing that the combination of paradigmatic radiation sensitizers with radiation quantitatively alters the induction of certain predefined mechanisms of cell death (Fig. 10.7). [Pg.184]
