It does not matter, from the fundamental point of view, how the pressure drop is generated in the filter. In the case of the centrifugal filters there is an additional phenomenon of the mass forces acting on the Hquid within the cake. The conventional filtration theory must be amended to include this effect (2).

The commercial production of mitomycin involves the preparation of mitomycin-containing broths by culturing a mitomycin-producing organism, e.g. Streptomyces caespitosus, in suitable media as described at length In the literature. At the end of the fermentation cycle the whole broth is usually centrifuged, filtered or otherwise treated to separate the solids (mycelia) from the supernatant which contains substantially all of the antibiotic activity. 

This unit is good to pre-thicken feed to centrifugal filters and similar applications. One cyclone may satisfy a requirement, or the units can be arranged in parallel for large capacities or in series for removal of extreme fines. See Figure 4-53 for a counter-current wash system. Solids as small as 10 microns can be separated. 

Ensure that the actual instrument configuration conforms to what is written under Experimental supplier, models, modifications, consumables (HPLC or GC columns, gaskets, etc.), and software for the main instrument, peripherals (injectors, integrators, computers, printers, plotters, etc.), and ancillary equipment (vortexer, dispensers, balances, centrifuges, filters, tubing, etc.). 

Weigh 50 g of the potato sample into a 250-mL centrifuge vial and homogenize with 100 mL of acetone. After centrifugation at 1500-2000 rpm for 5 min, decant the supernatant over filter paper filled with Celite and collect in a 500-mL evaporation flask. Repeat extraction with 80 mL of acetone, centrifuge, filter and combine the extracts. Rinse Celite with 20 mL of acetone into the combined filtrates. 

Centrifugal filters use centrifugal force to drive the filtrate through the filter cake. The equipment used is described in the next section.

It is convenient to classify centrifugal filters into two broad classes, depending on how the solids are removed fixed bed or moving bed. 

The problem of crystallizing dextrose from starch conversion liquor in a form which would allow separation from the viscous mother liquor was attacked in the latter part of the nineteenth century by various workers. The most successful efforts were made by Behr in 1881.3 Corn starch was hydrolyzed at low concentration, the refined product was concentrated, seeded with a very small proportion of pure anhydrous dextrose crystals, and it was then allowed to stand without agitation in heated rooms. Care was taken to exclude any traces of dextrose hydrate crystals. After several days the crystalline magma was separated and washed in centrifugal filters. Another method used a semi-preparative ODS column for the prefractionation of wine anthocyanins. Alcohol from wine samples was evaporated at ambient temperature, the aquoeus rest was centrifuged, filtered and applied into an ODS column (230 X 15 mm i.d.). The material eluted was loaded onto the column once more subsequently, the column was washed with 350 mL of 20 mM KH2PO4 (pH 7.4) and then with 350 mL of 20 him potassium phosphate buffer (pH 7.4) supplemented with 1 mM imidazole. PAMO-P3 was eluted with 100 mL of 50 mM tris-HCl (pH 7.4) containing 200 mM imidazole. In total, 25 mL of yellow eluate was collected and concentrated to 12.5 mL by centrifuge filters (Amicon, MWCO 10000). 

Fifty-microlitre aliquots of standards, QC material or neat CSF are injected directly onto the F1PLC system. Blood-contaminated CSF should be filtered using a 10,000-molecular-weight (MW) centrifugal filtration system (e.g. Millipore Ultrafree-MC centrifugal filters) prior to injection.
