Benzene derivatives substitution

During their work on the arylation of aromatic compounds by substitution, Fujiwara, et al. observed biaryl formation when aromatic compounds were placed in the presence of olefin-palladium complexes and silver nitrate.80 Developing this reaction as a method for biphenyl synthesis, these authors showed that the more stable the olefin-palladium complex was, the lower the yield. Ethylene dichloropalladium proved to be the best choice, when used with silver nitrate. However, the reaction required stoichiometric amounts of both catalysts (Scheme 10.47). Benzene derivatives substituted by electron-donating or -withdrawing groups reacted as well, but a mixture of regioisomers was produced, except for nitrobenzene, which only gave m,m -dinitrobiphenyl. 

The variety of synthetic possibilities which may be envisaged should result in the rapid development of these reactions. Note that using a different principle related to the aromatic trimerization of alkynes by cobalt (Heck, 1969), benzene derivatives substituted in well defined positions (CXXIII) may be obtained in moderate yields (13-59%) by reaction of cobaltocy-clopentadienyls (CXXII) (Wakatsuki et al, 1974) with alkynes according to Scheme 12.

A point in case is provided by the bromination of various monosubstituted benzene derivatives it was realized that substituents with atoms carrying free electron pairs bonded directly to the benzene ring (OH, NH2, etc) gave 0- and p-substituted benzene derivatives. Furthermore, in all cases except of the halogen atoms the reaction rates were higher than with unsubstituted benzene. On the other hand, substituents with double bonds in conjugation with the benzene ring (NO2, CHO, etc.) decreased reaction rates and provided m-substituted benzene derivatives. 

The course of aromatic substitution has been placed on a more scientific basis by the following rules of Hammick and Illingworth (jfour. Chem. Soc., 930. 2358), If a monosubstituted benzene derivative has the formula CgHsXY, where X is the atom joined to the benzene ring and Y is an atom or group of atoms attached to X, then —... 

A brief account of aromatic substitution may be usefully given here as it will assist the student in predicting the orientation of disubstituted benzene derivatives produced in the different substitution reactions. For the nitration of nitrobenzene the substance must be heated with a mixture of fuming nitric acid and concentrated sulphuric acid the product is largely ni-dinitrobenzene (about 90 per cent.), accompanied by a little o-dinitrobenzene (about 5 per cent.) which is eliminated in the recrystallisation process. On the other hand phenol can be easily nitrated with dilute nitric acid to yield a mixture of ortho and para nitrophenols. It may be said, therefore, that orientation is meta with the... 

If, on the other hand, the encounter pair were an oriented structure, positional selectivity could be retained for a different reason and in a different quantitative sense. Thus, a monosubstituted benzene derivative in which the substituent was sufficiently powerfully activating would react with the electrophile to give three different encounter pairs two of these would more readily proceed to the substitution products than to the starting materials, whilst the third might more readily break up than go to products. In the limit the first two would be giving substitution at the encounter rate and, in the absence of steric effects, products in the statistical ratio whilst the third would not. If we consider particular cases, there is nothing in the rather inadequate data available to discourage the view that, for example, in the cases of toluene or phenol, which in sulphuric acid are nitrated at or near the encounter rate, the... 

Butyne trimerizes in the presence of aluminum chloride to give hexamethyl Dewar-benzene (W. Schafer, 1967). Its irradiation leads not only to aromatization but also to hexa-methylprismane (D.M. Lemal, 1966). Highly substituted prlsmanes may also be obtained from the corresponding benzene derivatives by irradiation with 254 nm light. The rather stable prismane itself was synthesized via another hydrocarbon, namely benzvalene, a labile molecule (T. J. Katz, 1971, 1972). 

Piperazinothiazoies (2) were obtained by such a replacement reaction, Cu powder being used as catalyst (25. 26). 2-Piperidinothiazoles are obtained in a similar way (Scheme 2) (27). This catalytic reaction has been postulated in the case of benzene derivatives as a nucleophilic substitution on the copper-complexed halide in which the halogen possesses a positive character by coordination (29). For heterocyclic compounds the coordination probably occurs on the ring nitrogen. 

The two substituted carbons are connected by a double bond in one structure but by a single bond in the other Because no such cases of isomerism m benzene derivatives were known and none could be found Kekule suggested that two isomeric structures could exist but mterconverted too rapidly to be separated... 

The name phenylene o-, m-, or p-) is retained for the radical —C5H4—. Bivalent radicals formed from substituted benzene derivatives and having the free valences at ring atoms are named as substituted phenylene radicals, with the carbon atoms having the free valences being numbered 1,2-, 1,3-, or 1,4-, as appropriate.

Calixarenes (from the Latin ca/ x) may be understood as artificial receptor analogues of the natural cyclodextrins (96,97). In its prototypical form they feature a macrocycHc metacyclophane framework bearing protonizable hydroxy groups made from condensation of -substituted phenols with formaldehyde (Fig. 15b). Dependent on the ring size, benzene derivatives are the substrates most commonly included into the calix cavity (98), but other interesting substrates such as C q have also been accommodated (Fig. 8c) (45). 

Electrophilic substitution of thiophene occurs largely at the 2-position and the reactivity of the ring is greater than that of benzene. 3-Substituted derivatives are generally prepared by indirect means or through ring cyclization reactions. 

Nucleophilic Substitutions of Benzene Derivatives. Benzene itself does not normally react with nucleophiles such as haUde ions, cyanide, hydroxide, or alkoxides (7). However, aromatic rings containing one or more electron-withdrawing groups, usually halogen, react with nucleophiles to give substitution products. An example of this type of reaction is the industrial conversion of chlorobenzene to phenol with sodium hydroxide at 400°C (8). 

Reactions of acetylene and iron carbonyls can yield benzene derivatives, quinones, cyclopentadienes, and a variety of heterocycHc compounds. The cyclization reaction is useful for preparing substituted benzenes. The reaction of / fZ-butylacetylene in the presence of Co2(CO)g as the catalyst yields l,2,4-tri-/ f2 butylbenzene (142). The reaction of Fe(CO) and diphenylacetylene yields no less than seven different species. A cyclobutadiene derivative [31811 -56-0] is the most important (143—145). 

A -Amino- and A-substituted amino-pyrroles readily undergo Diels-Alder additions and add to activated alkynes at room temperature. The resulting azanorbornadienes extrude A-aminonitrenes and this forms the basis of an unusual synthesis of benzene derivatives (81S753,81TL3347). It has been found that ethyl/3-phenylsulfonylpropiolate (135) is a superior dienophile to DMAD (Scheme 50). 

As appropriately substituted o-disubstituted benzene derivatives are feadily available, this procedure has found widespread application in the synthesis of benzo-fused flve-membered heterocycles. Examples abound in the various chapters in these volumes and the following few examples illustrate the general trend. 

Several alkenes are converted to aziridines by treating with oxaziridine (52) at elevated temperatures. Styrene, a-methylstyrene and their derivatives substituted in the benzene ring react smoothly, and so do 1,1-diphenylethylene, indene and acrylonitrile (74KGS1629). 

In the early work, benzene formed the basis of a variety of multi-armed structures. Analogs bearing from 2—6 arms were prepared and compared for cation binding ability. The only indication of mode of synthesis for the hexa-substituted benzene derivative is that it was obtained on reaction of benzene-hexakis(methanethiol) and l-bromo-3,6,9-trioxatridecane . The reaction is illustrated in Eq. (7.6), below, devoid of reaction conditions and yields which were not specified. 

Phenol and anisole are among the commonly encountered benzene derivatives listed in Table 11.1. Electrophilic aromatic substitution in phenol is discussed in more detail in Section 24.8. 

The reduction of the C— Br and C—1 group moments from 1.10 and 0.90 in bromo- and iodo-benzene to about 0.80 and 0.50 in 2-bromo- and 2-iodo-thiophene has been ascribed to the larger weight of resonance forms such as (8) and (9) in the thiophene series. The chlorine, nuclear, quadrupole, resonance frequencies of chloro-substituted thiophenes are much higher than those of the corresponding benzene derivatives. This has been ascribed to a relayed inductive effect originating in the polarity of the C—S o-bond in thiophenes. The refractive indices, densities, and surface tension of thiophene, alkyl- and halo-thiophenes, and of some other derivatives have been... 

Charton has recently examined substituent effects in the ortho position in benzene derivatives and in the a-position in pyridines, quinolines, and isoquinolines. He concludes that, in benzene derivatives, the effects in the ortho position are proportional to the effects in the para position op). However, he finds that effects of a-sub-stituents on reactions involving the sp lone pair of the nitrogen atoms in pyridine, quinoline, and isoquinoline are approximately proportional to CT -values, or possibly to inductive effects (Taft s a ). He also notes that the effects of substituents on proton-deuterium exchange in the ortho position of substituted benzenes are comparable to the effects of the same substituents in the a-position of the heterocycles. 

Several ad hoc studies and discussions in recent years have been centered around the mechanism of aromatic substitution in nitro-activated benzene derivatives. The subject has been reviewed authoritatively. ...
