The soluble tryptic peptides of 130 mg a chain of Hb-St. Claude were separated on 0.9 x 60 cm columns of Chromobead resin type P (Technlcon Instruments, Dowex 50-X4) at 37°C using the procedure described earlier (16). The method uses a gradient of volatile pyrldlne-acetlc acid buffers of differing molarities and pH as follows first gradient, 666 ml 0.1 M, pH 3.1, and 333 ml 1.0 M, pH 5.0 and second gradient, 166 wl 1.0 M, pH 5.0, and 332 ml 2.0 M, pH 5.0. The amino acid composition of Isolated fragments was determined with a Splnco model 121 automated amino acid analyzer (Beckman Instruments)... [Pg.37]

In the Beckman instrument, change in conductivity is used (28). The instrument uses 14pi for the test but samples from a cup which needs to contain at least 100 pi of serim. [Pg.122]

The Beckman instrument suffers from certain faults. First, the standard for urea must contain the proper amount of saline or false values are obtained. Second, urine cannot be analyzed... [Pg.122]

Table I compares the measured values of extinction coefficients with the values calculated from theory at wavelengths of 25U, 280 and 350 nm. The lower values of extinction coefficients for the larger particles measured, using the online detector compared to the Beckman instrument, are due to the optical effects discussed above. The disagreement between the extinction coefficients calculated from Mie theory for non-absorbing spheres and the measured values from the Beckman instrument is very significant, particularly at 2 k and 280 nm. The surface concentration of adsorbed albumin was then determined by comparing the radioactivity of an aliquot of solution of known protein concentration as described previously [3.4]. For this purpose, a Beckman Instrument (Palo Alto, CA) y-counter was used. [Pg.171]

One of the best-known commercial instruments developed for organic carbon determinations is the Beckman total carbon analyser, which utilises an analysis scheme developed by Van Hall and co-workers [57,99]. This instrument works reasonably well in fresh water. It has become a standard instrument in pollution control and water treatment [103]. The Beckman instrument has not worked as satisfactorily for seawater because of the latter s high carbonate and low organic content. [Pg.496]

Aroxmd the same time, Beckman Instruments (now Beckman Coulter) had begun an array-based product development program focused on the use of modified plastics. Silzel and coworkers (1998) and Matson et al. (2001) of Beckman Coulter were among the first to pursue printing of antibodies onto a plastic surface in a microarray format. Silzel et al. immobilized biotinylated monoclonal antibodies onto an avidin-coated polystyrene surface and performed micro-ELISA-based isotyping of IgG species. Matson et al. [Pg.70]

Figure 1. Kinetics of K efflux from Chlorella under Og stress. External K measured with cation-specific electrode (Beckman Instruments, Fullerton, model 39137) with 10" cells/ml suspended in a 10 mM Tris-Cl, ImM CaCU, pH 9 solution (10 ml, total volume). Control efflux (0—0) is linear after 20 min (denoted as 0 time here). The addition of Og ( — ) for 30 min and continuous Og (O—O), 26 fimoles/liter air flow (25 cc/min) are shown. The electrode output is amplified by an electrometer coupled to an antilog converter (12,13). Figure 1. Kinetics of K efflux from Chlorella under Og stress. Edman and Begg s publication in 1967 initiated attempts at many places to construct a similar instrument. At the end of 1969 an instrument became commercially available developed by Beckman Instruments, Palo Alto, Calif. This machine, termed Protein Peptide Sequencer , generally follows the specifications of Edman s prototype but incorporates some novel solutions for technical problems. Such an instrument is at the disposal of the authors and shall be described in the next chapter. [Pg.11]
