This section discusses treatment of the water samples in preparation for instrumental analysis after they have been received, archived and stored in the laboratory. Two major panels of experts publish guidelines for the treatment of HIV-infected individuals. Although the recommendations are quite similar, slight differences do exist between the Department of Health and Human Services (DHHS) Guidelines2 and the International AIDS Society-USA (IAS-USA) Panel Recommendations.4 The DHHS Guidelines are updated every 6 months and current and archived versions are available online at www.aidsinfo.nih.gov. This presentation demonstrates that a small minicomputer can be used to provide a full range of functions for collection and interactive reduction of data from a size-exclusion liquid chromatograph. A number of different users have collected in excess of 5000 chromatograms using this equipment. The experience gained with this system has influenced our approach to the automation of other analytical instruments. The heat-induced retrieval protocol for extraction of formaldehyde-modified DNA from FFPE tissue sections provides a simple and effective method of DNA extraction from archival tissue samples.25,45 Based on PCR using three primer pairs ranging from 152-541 bp and a real time KTC-PCR analysis, the heat-induced retrieval protocol yields a better quality and quantity of DNA samples extracted from FFPE tissue sections than conventional methods of extraction.24 In addition, this heating protocol may provide an alternative approach for DNA extraction in some cases such as a recent publication by Ferrari et al. mentioned above.34... [Pg.54]
