Apoptotic cell death is regulated by NO and ONOO" in several cell types including myeloid-derived leukocytes such as neutrophils. The biological effects of NO and ONOO" have been recently reviewed [68]. GEA3162 is able to promote apoptotic cell death in human neutrophils in a caspase-dependent manner [69]. Antisense disruption of Mcl-l protein levels in human neutrophils can decrease the levels of this survival factor in control or GM-CSF-untreated cells (fig. 2). These decreases in protein levels occur in the absence of changes in the levels of other members of the Bcl-2 family. As Mcl-l levels are decreased by antisense treatment, there is a corresponding increase in the cellular levels of the active form of caspase 3 (fig. 2).
