A wide selection of monoclonal and polyclonal anti-Ca -ATPase antibodies have become available in recent years. Studies with these antibodies defined the localization of Ca " -ATPase in the sarcoplasmic reticulum of developing and mature skeletal muscles [60,262-270] and established a pattern of cross reactivity with various Ca -ATPase isoenzymes in the sarco(endo)plasmic reticulum [270-286] and in the plasma membrane [284,287-290] of skeletal, cardiac and smooth muscles. Antibodies have also proved useful in the quantitation of Ca -ATPase, both in muscles of diverse fiber types [291-294] and in COS-1 cells transfected with Ca -ATPase cDNA [97,103,126,127,129,215],...

At an early stage in product development, there must be some material made available for use as ELISA reagents. ELISA requires a standard for quantitation and specific antibodies to the drug protein of interest. If these are not already available, either commercially or internally, then time, effort, and expense must be reserved for reagent development as well as assay development activities. The specific antibody is the key reagent in an ELISA. The antibody defines the specificity and sensitivity of the assay. To date there is no successful substitution for the routine production of specific antibodies by immunization of an animal with the antigen target of interest.

Two other monoclonal antibodies define CA2 and CA3 on N-linked oligosaccharides, but these are less characterized than CAl [83]. Competition studies suggest that CA2 may recognize GlcNAc-6-S04 residues. CA3 shows little preference for inhibition by different sugars.
