The homogeneity determination of the bacteria in the materials is performed by viable count followed by statistical evaluation of the cormts of sub-samples from the same capsule solution and of total counts of different capsules of one batch. An example for the homogeneity determination for a batch of capsules containing Enterococcus faecium is also presented in (Janning et al. 1995).

The tests for microbial limits and recommendations for microbial quality criteria of raw materials, excipients, drug substances, and pharmaceutical products have been established in pharmacopoeial compendia for over 30 years. These tests are listed in the USP 24 Chapter (61) Microbial Limits Tests and in the Ph. Eur. 3rd ed.. Biological Tests 2.6.12 and 2.6.13, Microbial Contamination of Products Not Required to Comply with the Test for Sterility (total viable count, tests for specified microorganisms) and the JP XIII 30 Microbial Limit Test. 

Total viable counts (TVCs) were immediately carried out on duplicate controls with no biocide added. The slurries were incubated at 37 °C for 4 hours and TVCs were performed at that time point and at several points over the following 6 weeks. Re-inoculations were carried out after 3 weeks. The resulting bacterial numbers were recorded as colony forming units (cfu)/ml of slurry. 

Sensory. Sensory analysis is one of the oldest accepted methods forjudging seafood quality and freshness. Although fairly accurate, it unfortunately requires trained, experienced panelists to attain accuracy. Perez-Villarreal (( u evaluators experienced in sensory analysis of fish to judge freshness and quality. Results of the study showed sensory scores were better correlated with microbiological analyses, especially total viable counts, than with chemical analyses such as trimethylamine or enine nucleotide decomposition (6).

Microbial limit tests, as described in USP 24-NF 19 (<61>) provide the information necessary to conduct bioburden counts by quantitative estimations of viable aerobic microorganisms in pharmaceutical articles, raw materials, and finished dosage forms. Consult the EP or JP for any differences in these methods from those of the USP. Note that USP (<61>) is currently under revision and that the next version will be fully harmonized between the USP, EP, and JP. The microbial limit test comprises the total aerobic bacterial (microbial) count (TAG), the total combined yeasts and molds count (TYMC), and tests for indicator organisms. Bioburden tests are also referred to as total viable counts. This test estimates the total number of viable (culturable) aerobic microorganisms in various items including parenteral UBPS. The total viable count is obtained by adding the counts from the TAC and the TYMC.

The results on Table 2 shows a Methylene Bisthiocyanate based biocide. The mechanism of action of this biocide is to block the transfer of electrons from primary cytochrome dehydrogenase, and thereby cause an uncoupling of oxidative phosphorylation . Here the Total ATP results initially increase on treatment with the biocide a small increase in Free ATP is also evident along with a reduction in viable counts. With time there is a reduction in Total and Free ATP along with a further reduction in the plate count results.

Wang et al. [44] used an EN equipped with MOX sensors together with support vector machine (SVM) and partial least squares (PLS) to predict the total viable counts in chilled pork samples. The achieved correlation coefficients for training and validation were close to 90 %, which suggested that the EN system could be used as a simple and rapid technique for absolving the task.
