Validation background

Whereas chromatograms obtained as background controls for many agricultural crops vary with the natural chemical composition and degree of ripeness, chromatograms for pollen can become further complicated by the unpredictable foraging habits of honey bees (Apis mellifera L.), and the question of what constitutes a valid background control sample. 

Current knowledge permits a more definitive description of the enzymes than was possible in EIC-I however, the fundamental chemistry described therein provides valuable context and valid background for the present account. The simplicity mentioned above exists for various kinds of samples that meet the fundamental requirements laid down in Chapters 6 and 7. Examples are a thin film on a suitable substrate, a sample dissolved at low concentration in a solvent transparent to x-rays, or a sample uniformly dispersed in a similarly transparent medium. In all cases, scattered x-rays should be at a minimum to keep the background low. From the point of view taken here, a trace is thus regarded as a major constituent in a sample if sensible absorption and enhancement effects are absent—if, that is, Equation 7-3 is valid.

In this paper, we discuss studies based on comparison with background measurements that may have a skew distribution. We discuss below the design of such a study. The design is intended to insure that the model for the comparison is valid and that the amount of skewness is minimized. Subsequently, we present a statistical method for the comparison of the background measurements with the largest of the measurements from the suspected region. This method, which is based on the use of power transformations to achieve normality, is original in that it takes into account estimation of the transformation from the data. 

The purpose of this article is to clarify the assessment of residue analytical methods in the context of Directive 91/414/EEC. After discussing the legal and historical background, requirements for enforcement methods as well as data generation methods are reviewed. Finally, an outlook over further developments in the assessment and validation of analytical methods is provided.
