Whereas commercial systems usually employ stirred mini vessels, laboratory-developed apparatus may be operated in flow-through mode. For mini-vessels (vials, wells), the titer plate format is typical and widely accepted. 

Permeability-pH profiles, log Pe - pH curves in arhficial membrane models (log Pjpp - pH in cehular models), generally have sigmoidal shape, similar to that of log Dod - pH cf. Fig. 3.1). However, one feature is unique to permeabihty profiles the upper horizontal part of the sigmoidal curves may be verhcally depressed, due to the drug transport resistance arising from the aqueous boundary layer (ABL) adjacent to the two sides of the membrane barrier. Hence, the true membrane contribution to transport may be obscured when water is the rate-limiting resistance to transport. This is especially true if sparingly soluble molecules are considered and if the solutions on either or both sides of the membrane barrier are poorly stirred (often a problem with 96-well microhter plate formats). 

An automated log P workstation using a shake-flask method and robotic liquid handling in 96-well plate format is commercially available [30]. The system is equipped with a diode-array spectrophotometer and equimolar nitrogen detector. 

Assay conditions as determined by ASSAY VALIDATION are performed in the chosen PLATE FORMAT with an acceptable signal to background ratio as described by the Z factor.

We have developed chemiluminescent immunoenzymatic assays for (3-ago-nist drugs in the 96-well-microtiter-plate format. Such competitive assays have been used for determination of clenbuterol and of the overall content of p-agonist drugs in the sample. They matched the standard requirements of precision and accuracy, and were more sensitive compared to the conventional colorimetric methods. Moreover, CL detection was very rapid, making these assays suitable for screening analysis. 

The simplest technique is the use of the 96-well collection plate format (analogous to the format used in SPE) in conjunction with a liquid handling robotic system it follows the same principle as bulk scale LLE. However, immobilization of the aqueous plasma sample on an inert solid support medium packed in a cartridge or in the individual wells of a 96-well plate and percolating a water-immiscible organic solvent to extract the analyte from this medium evoked significant enthusiasm from the pharmaceutical industry.
