Ketorolac, which appears to have a greater analgesic than anti-inflammatory or antipyretic activity, is widely used to treat postoperative as well as chronic pain. The tromethamine salt possesses sufficient water solubility to allow for parenteral administration. When administered orally or intramuscularly, ketorolac is rapidly and well absorbed, with peak plasma concentrations attained between 30 min and 50 min. In doses of 10-30 mg intramuscularly it is... [Pg.137]

Ketorolac (administered as the tromethamine salt Toradol, Ultram) has been used as a short-term alternative (less than 5 days) to opioids for the treatment of moderate to severe pain and is administered intramnscnlarly, intravenously, or orally. Unlike opioids, tolerance, withdrawal, and respiratory depression do not occur. Like other NSAIDs, aspirin sensitivity is a contraindication to the nse of ketorolac. Typical doses are 30 to 60 mg (intramnscnlar), 15 to 30 mg (intravenous), and 5 to 30 mg (oral). Ketorolac is used widely in postoperative patients, but it should not be used for routine obstetric analgesia. Topical (ophthalmic) ketorolac is FDA approved for the treatment of seasonal allergic conjunctivitis and postoperative ocular inflammation after cataract extraction. [Pg.375]

Tromethamine salt, C-jH NO, Lu to lyse, Prostin F, Alpha Injectable. White or off-white cystalline powder. Readily sol in water to at least 200 mg/ml. [Pg.1252]

The stability in the solid state of the free acid and three salts of a prostaglandin derivative were compared on samples stored at 33°C, protected from light. After 2 months, > 10% of the acid, > 50% of the sodium salt, < 2% of the potassiuni salt and 2-3% of the Tris (= tromethamine) salt were decomposed. The surprisingly large differences could be correlated neither with the water solubility nor with the melting point. ... [Pg.613]

Fosfomydn Tromethamine. Fosphomycin trometha-mine (Monurol) is a phosphonic acid epoxide derivative that was initially isolated from fermentations of Streptomyces spp. The structure of the drug is shown below. Making the tromelhuminc salt greatly expanded the therapeutic utility of this antibucleriul because water solubility increased enough to allow oral administration. [Pg.364]

Usually companies do not investigate other salt-forming species other than those shown in Table 3.8. However, Gu and Strickley (1987) have described the physical properties of the tris-(hydroxymethyl)aminomethane (THAM) salts of four analgesic/antiinflammatory agents with sodium salts and free acids. They concluded that the THAM salts had superior hygroscopicity properties compared to the sodium salts and did not show any loss in aqueous solubility or intrinsic dissolution rate. The one exception to this was the THAM salt of naproxen. Ketoralac is now marketed as the tromethamine or THAM salt. [Pg.54]
