Thymidine uptake

The most convenient repair assay is thymidine uptake into Go lymphocytes (Evans and Norman, 1968). Since there is little semiconservative DNA synthesis in such cells, and what synthesis there is may be further reduced by the addition of hydroxyurea, background incorporation is essentially zero and all the counts incorporated into an acid-insoluble cell fraction can be assumed to result from repair synthesis. This method is rapid and convenient, but by itself it provides no information as to the number or size of repaired regions obviously, many repaired regions with a few base insertions or a few repaired regions with many base insertions would give similar results in terms of thymidine uptake. Nevertheless, for a quick measure of repair after insult by some suspected DNA-damaging agent, the method is ideal. [Pg.155]

Thymidine Uptake Studies. Tritiated thymidine (52 mCi/ anole 0.1 /rCi/ml) was added to cells for 60 min at 37°C. Cells were then washed with PBS, incubated at 4°C for 15 min in the presence of ice cold 5% TCA, rinsed with TCA and scraped from the flasks with a rubber policeman. Cells were again washed with PBS, and solubilized in 0.1N NaOH overnight. Aliquots were assayed for protein (62) and radioactivity (scintillation fluid 100 ml Biosolve (Beckman, Fullerton, CA.), 7g of PPO and 0.6 g of POPOP per liter of toluene). [Pg.248]

A depressed uptake of deoxyuridine brought about by a deficiency of vitamin B12 can be reversed as reflected by a reduction in labeled thymidine uptake by the addition of as little as 1 xg of the vitamin. Folic acid added to a concentration of 50 p.g/ml of the culture will correct abnormal results due to folate deficiency and may partially correct abnormal results due to vitamin B12 deficiency. Organic and aqueous feverfew powdered leaf extracts were found to inhibit IL-1-induced prostaglandin E2 release from synovial cells, IL-2-induced thymidine uptake by lymphoblasts, and mitogen-induced uptake of thymidine by peripheral blood mononuclear cells (PBMCs) (23). Parthenolide also inhibited thymidine uptake by PBMCs. Both parthenolide and the extracts were cytotoxic to the PBMCs and synovial cells thus, the anti-inflammatory effects of feverfew may be secondary to cytotoxicity. These results reflect those of previous researchers who found parthenolide and other sesquiterpene lactones to be cytotoxic to cultures of human fibroblasts, human laryngeal carcinoma cells, and human cells transformed with simian virus 40 (24). [Pg.118]

In an attempt to define whether chalone acts at the phase (Bullough s antephase) or earlier in the cell cycle, Baden and Sviokla (B3) did experiments using both the mitotic arrest technique with Colcemid and scintillation counting of tritiated thymidine uptake into DNA isolated from epidermis exposed to chalone.
