For kinetic experiments, when the regular recovery experiment is completed (when using a real drug product, not from spiked experiments), the solution that is left over is set aside for time t (usually 24 hours). After time t, the solution is re-shaken by hand and then re-injected to determine assay value. [Pg.715]

For a thermodynamic experiment, it is necessary to perform the regular recovery experiment using a real drug product with more volume of sample solvent. For example, if the procedure is stated to extract the content of a drug product with 50 mL of solvent, then use 75 mL and/or 100 mL for this experiment. [Pg.715]

The results from these two experiments (kinetic and thermodynamic) will show whether the regular extraction procedure is complete or not. Most hkely, for modified-release drug products, time is essential (higher recovery over time, but watch out for solution stabihty ). The change in volume will have an impact if the solubihty of an API is on the border of the solubility limit in that particular sample preparation solvent (in the presence of excipients). If the latter is the case, then the procedure should be modified to extract with higher volume of sample preparation solvent and/or change the pH or composition of the solvent. [Pg.715]

Lastly, to really prove that the sample preparation procedure or the recovery procedure is completely extracting your API and degradation products, utilize homogenization (refer to section on automation) instead of mechanical shaking and/or sonication.
