Test Part Four

Such an approach was trialled in the late 1990s as part of an initiative by the Environment Agency in the UK. It led to proposals for a quality control scheme for use within the Agency s Direct Toxicity Assessment programme for effluent assessment and control. The trial entailed an interlaboratory ring-test of four test methods using two toxicants (3,4-dichloroaniline and zinc sulphate) from which accuracy and precision criteria were estimated, as described below.

The turbine undergoes three basic tests, these are hydrostatic, mechanical, and performance. Hydrostatic tests are to be conducted on pressure-containing parts with water at least one-and-a-half times the maximum operating pressure. The mechanical run tests are to be conducted for at least a period of four hours at maximum continuous speed. This test is usually done at no-load conditions. It checks out the bearing performance and vibration levels as well as overall mechanical operability. It is suggested that the user have a representative at this test to tape record as much of the data as possible. The data are helpful in further evaluation of the unit or can be used as base-line data. Performance tests should be conducted at maximum power with normal fuel composition. The tests should be conducted in accordance with ASME PTC-22, which is described in more detail in Chapter 20. 

Operator training was planned to take place four months prior to the first heat-up. Half of the operators were to be on the payroll five months, and the other half for three months. As part of the training, each operator was to have spent four weeks in the plant inspecting construction, becoming familiar with the equipment, and helping with hydrostatic testing and internal cleaning.

We initially tested Candida antarctica lipase using imidazolium salt as solvent because CAL was found to be the best enzyme to resolve our model substrate 5-phenyl-l-penten-3-ol (la) the acylation rate was strongly dependent on the anionic part of the solvents. The best results were recorded when [bmim][BF4] was employed as the solvent, and the reaction rate was nearly equal to that of the reference reaction in diisopropyl ether. The second choice of solvent was [bmim][PFg]. On the contrary, a significant drop in the reaction rate was obtained when the reaction was carried out in TFA salt or OTf salt. From these results, we concluded that BF4 salt and PFg salt were suitable solvents for the present lipase-catalyzed reaction. Acylation of la was accomplished by these four enzymes Candida antarctica lipase, lipase QL from Alcaligenes, Lipase PS from Burkholderia cepacia and Candida rugosa lipase. In contrast, no reaction took place when PPL or PLE was used as catalyst in this solvent system. These results were established in March 2000 but we encountered a serious problem in that the results were significantly dependent on the lot of the ILs that we prepared ourselves. The problem was very serious because sometimes the reaction did not proceed at all. So we attempted to purify the ILs and established a very successful procedure (Fig. 3) the salt was first washed with a mixed solvent of hexane and ethyl acetate (2 1 or 4 1), treated with activated charcoal and passed into activated alumina neutral type I as an acetone solution.
