Target types

Figure 11-69 describes a typical central system. Either water or direct-expansion refrigerant coils or air washers may be used for cooling. Steam or hot-water coils are available for heating. Humidification may be provided by target-type water nozzles, pan humidifiers, air washers, or sprayed coils. Air cleaning is usually provided by cleanable or throwaway filters. Central-station air-conditioning units in capacities up to about 50,000 cu ft/min are available in prefabricated units. 

One target type for which the molecular mechanism of efficacy has been partly elucidated is the G-protein-coupled receptor (GPCR). It is known that activation of GPCRs leads to an interaction of the receptor with separate membrane G-proteins to cause dissociation of the G-protein subunits and subsequent activation of effectors (see Chapter 2). For the purposes of binding, this process can lead to an aberration in the binding reaction as perceived in experimental binding studies. The truth most likely lies somewhere in between. Bender [67] published the most quantitative study to date on the success of HTS at Novartis. Several conclusions could be drawn. Particular target types and assay technologies have a great impact on screening success, and this was not always correlated to the number of identifying hits in the HTS runs. For assay formats used a minimum of five times, LC/MS readouts succeed 83% of the time, followed by FP assays, which succeed in 72% of the cases. If a type has the same name as its package, then using that name as a stereotype on a target type means to import the package, substituting the target type. 

Focus on one type at a time and ask yourself, At any one moment, what information would we want to record about this type of object Write this information as attributes and associations sourced at this type. For each one, ask what the type of that information is, giving the target type. Don t confuse static information with actions actions abstract interactions, whereas attributes represent information known before and/or after the actions. For example ... 

An attribute, like a link, is a read-only query. The main difference is that it is shown as text rather thanpictorially. In theory, the two forms are interchangeable, but in practice the attribute form is used when the target type is defined in some other body of work. The first level of genotoxicity testing likely to be used in the near future is hit profiling. For numerical reasons, hits from a discovery campaign can be considered alongside smaller complete libraries of natural products and specialist collections enriched for particular target types (inhibitors of kinases, proteases, HDACs, etc.). This review is organized primarily on the basis of the electrophiles used, with a view to allowing easy access to the information.
