Sumner value

The value of 483,000 is in the original publication but was misquoted by both Sumner (1) and Varner (3) as 473,000. Moreover, the diffusion constant was determined in the presence of sodium sulfite but the sedimentation constant was not. The main component in phosphate buffer was found to have a sedimentation constant of 18 5 at 20°. 

The isoelectric point, determined by Sumner and Hand (50) to have a value of 5.0-5.1, has been redetermined by the electrofocusing technique (43). The value obtained was 4.8. The solubility is extremely small at this pH, but the urease can be located by its enzymic action. It is a point of interest that the solubility of the isoelectric species increases spectacularly if the sulfhydryl groups are substituted with A-ethylmaleimide (NEM) (43). 

D. E. Koshland and J. T. Edsall have recorded their own experiences in early protein research. Koshland stresses the value of his induced fit theory,192 while Edsall describes his long career at Harvard after two years with Hopkins at Cambridge.193 Fruton has also described the work of T. B. Osborne (1859-1928), noted for his analyses of amino-acids from seed proteins.194 The idea that enzymes might be proteins was a matter of heated debate among chemists from about 1915, including arguments between Willstatter and James Sumner, who in 1926 isolated the enzyme urease and showed it to be a protein.195 However, while it is most important to emphasize protein chemistry, the contributions made to protein science by physics...

Sumner and his co-workers (29) get about the same activity for beef and horse liver catalases for which we find is 3 x 10 Af sec. Thus the value of Beers and Sizer is definitely too low for a pure beef liver catalase. 

This represents a revision of the original value of 248,000 given by Sumner and Gralen.

Sumner also showed that the presence of an electrolyte can make a considerable impact on the difference between the internal pH and the pH in the liquor, and that this relationship is different for different fibres. For instance, in the case of cellulose the addition of sodium chloride reduces the difference between the internal and the external pH values, but the internal pH never exceeds that of the dyebath. The difference can be as much as 3 pH units when no electrolyte is present. The presence of 1 mol.T of NaCl, however, reduces this difference to about 0.2 pH units. In the presence of anionic dyes, this difference becomes even larger. However, a study of nylon type fibres reveals that, in the absence of dye, the internal and external pH values intersect at the isoelectric point of the fibre. When the fibre carries a positive charge, the internal pH is always higher than that in the dyebath, while after the isoelectric point it is lower than the external pH.
