Strategic aspects

This section sets forth an EOP development plan that should be fulfilled before the start of the actual writing of any guidelines or plant specific procedures. It also contains a discussion of the role of the regulatory body and the required supporting role of the utility. The following sections provide guidance on how to produce a consistent package of technically correct plant specific EOPs. 

Strategic decisions on the principles to be applied in the EOF development/upgrade programme involve the items listed below. The basis for these decisions has been discussed in Section 2. The recommended state of the art approach is as follows  

EOPs should be symptom based or state based  

EOPs should be consistent with the design basis of the plant. EOPs may impact the final safety analysis report (FSAR), limits and conditions/ technical specifications and other safety documents, since the scope of EOPs extends from expected plant transients to BDBAs  

EOPs should cope with all possible accident situations and provide guidance for a wide variety of equipment failures and operator errors.

The various elements of the error management program such as the development of high quality procedures and training and effective feedback and communications systems need to be supported by policies and standards to implement these policies. The development of these policies is an important strategic aspect of the implementation process. 

Flow cytometry (FCM) is widely used for exploring mechanism of action of compounds that compromise proliferation since it is rapid, accurate and usable for any cellular context [5], In this chapter we want to point out technical and strategic aspects of use of FCM for cell cycle studies of a putative anticancer agent. As an example we used Edotecarin, a topi inhibitor, firstly evaluating proliferation outcome and classical DNA content analysis by propidium iodide, and then since the compound treatment produced cell cycle perturbation difficult to interprete, a two-parametric analysis by 5-bromo-deoxyuridine (BrdU) was applied for separating cell cycle phases. Moreover we put our efforts into identifing specific cell cycle arrest not easily demonstrable by previously described methods, through the use of in vitro kinetics ( pulse and chase ). Finally, in vivo assessment of efficacy and biomarkers modulation after treatment was analyzed.

This chapter deals with the synthesis of naturally occurring molecules (or related models) and focuses on the construction of medium size carbocycles by ring-closing metathesis (RCM). We have arbitrarily chosen to organize this chapter by increasing ring size. Strategic aspects and potential problems are discussed.
