Spot quality

A scanner with two lasers for Cy3 and Cy5 labeling is fairly good enough for most of the microarray experiments. However, multiple lasers are necessary for simultaneous detection of all four nucleotide polymorphisms in chip-based SNPs detection. Besides, an extra third flurophore attached to a sequence that specifically binds to a linker region of the DNA spots could be used for spotting quality control. [Pg.349]

Of course, one must ask whether a combination of DMSO and betaine would be of even greater benefit for spot quality. Hessner et al. (2003a), in efforts to develop a tracking system to measure microarray performance, found that 1.5 M betaine in 3% DMSO provided the greatest probe retention on PLL slides. Quality Audit. Another important responsibiUty of quahty assurance is the audit function. Using the quahty audit as a tool, QA can monitor the operation of the manufacturing faciUty a toU, ie, contract, manufacturer or raw material suppHer to assure that written procedures are in place and that there is documentation to indicate the procedures are being followed. Properly executed audits allow QA to spot potential weakness in the quahty system that could allow errors to occur. Tests for quality of sealing of anodic coatings have become internationally standardised.
