Some practical considerations

Solvent extraction is generally employed in analysis to separate a solute (or solutes) of interest from substances which interfere in the ultimate quantitative analysis of the material sometimes the interfering solutes are extracted selectively. Solvent extraction is also used to concentrate a species which in aqueous solution is too dilute to be analysed. 

The choice of solvent for extraction is governed by the following considerations. 

A high distribution ratio for the solute and a low distribution ratio for undesirable impurities. 

Sufficiently low viscosity and sufficient density difference from the aqueous phase to avoid the formation of emulsions. 

Ease of recovery of solute from the solvent for subsequent analytical processing. Thus the b.p. of the solvent and the ease of stripping by chemical reagents merit attention when a choice is possible. 

Since not aU solvents used as mobile phases can be obtained in deuterated form, or are prohibitively expensive, the chromatographic conditions must be tailored to a mobile phase that can be substituted by a more NMR-compatible solvent 

Another feature that needs to be considered is the fact that the NMR experiment is a static experiment and hence no consecutive bond making and bond breaking will take place as in a flow situation. Therefore, is it important to have a correct selector-to-analyte ratio (S/A value) during the NMR measurements. Both the choice of solvent and the S/A ratio will affect the amount of complexed analyte, due to the equilibrium situation between the sorbent and the analyte. Therefore, care must be taken that there is no unwanted signal overlap of the analyte and selector signals. Since an equilibrium situation is involved, it is important to use pure solvents in order to prevent competing interactions with possible impurities present in the solvent. It is also important that the interaction with the sorbent does not result in a conformational change of the analyte and hence result in different intramolecular distances.

Some Practical Considerations in the Use of Salt Bridges. Salt bridges are most commonly used to diminish or stabilize the junction potential between solutions of different composition and to minimize cross-contamination between solutions. Lastly, the shape of the vessel does not appear to have an influence on the efficiency of the extraction, although some practical considerations such as the ease of filling the vessels needs to be taken into account.
