Serum albumin, bovine fibroblasts

Uveitis is a general term used to describe a group of syndromes that have, as a common feature, inflammation of the uveal tract. Experimental autoimmune uveoreti-nitis can be induced in many animals by a subretinal injection of bovine serum albumin or fibroblasts, although most studies have been done in rabbits. A number of nonsteroidal, anti-inflammatory proteins are known which may be valuable for long-term therapy of uveitis if delivered from encapsulated cells. Some anti-inflammatory factors that are believed to be promising for the treatment of uveitis diseases are inhibitors of inflammatory cytokines, such as antibodies or soluble receptors. [Pg.118]

Column Chromatography. Sepharose beads containing covalently linked gangliosides (0.2 ml packed volume) were placed into a pasteur pipette containing a small amount of glass wool. Columns were washed with HEM containing 50 ug/ml bovine serum albumin (3 ml). Interferon solutions in MEM-albumin (1 ml) were placed on the columns, which were eluted with MEM-albumin at a flow rate of no more than one drop per minute. Fractions of 1 ml were collected and interferon titers determined in each fraction after serial two-fold dilution. Columns onto which mouse fibroblast interferon had been loaded, were eluted with MEM-albumin first, then with 0.07 M N-acetylneuraminyl lactose at pH 2. [Pg.393]

BSA monomer, ovalbumin (chicken), P-lactoglobulin (bovine milk), serum albumin (human), carbonic anhydrase (bovine), L-glutamic dehydrogenase (bovine liver), a-chymotrypsin (bovine), a-chymotrypsinogen A (bovine), immunoglobulin (bovine milk), pepsin, trypsin (bovine), and heparin were from Sigma. RNase and lysozyme (egg white) were from Calbiochem. The recombinant human basic fibroblast... [Pg.115]

L929 mouse fibroblast cells in this device changed their morphology due to the forces exerted on them. A gradient was created by introducing a standard culture medium with fetal bovine serum (FBS) at one inlet and a serum-free medium at the other. FBS contains proteins, such as serum albumin, that are needed for cell survival, division and growth. The cell density and attachment depended on a combination of sheer stress and FBS concentration [28]. [Pg.299]

Since cell adhesion depends on the presence of certain proteins on a surface, protein density gradients have also been prepared and tested. Bovine serum albumin (BSA) is known to dexnease cell adhesion, while other proteins, for example fibronectin, enhance the interaction. A fibronectin concentration gradient saturated with BSA leads to the selective adhesion of fibroblasts to the fibronectin-coated r ons (Fig. 14) and to endothelial... [Pg.528]

CNTs transported proteins into cells in a similar maimer to that of DNA- and peptide-functionalized CNTs mentioned above [153,154], SWNTs non-covalently conjugated with various proteins such as streptavidine, cyc-tochrome c, protein A and bovin serum albumin were investigated for carriage into mammalian cells like HeLa, NIH-3T3 fibroblast, HL60 and Jurkats cells [173,174],... [Pg.185]

Radiolabeled [l- C]18 3n-3 was purchased from Perkin-Elmer Life Sciences (Boston, MA). The free acid form of 24 5n-3 and 24 6n-3 were generous gifts from A. Spector and H. Sprecher. Human skin fibroblasts from normal controls and patients with peroxisomal or mitochondrial disorders were received from the Mental Retardation Research Center of the Kennedy Krieger Institute. Cells at 90% confluence were incubated with 0.05 pCi albumin-bound [1- C] 18 3n-3 in Dul-becco s modified Eagle s minimum essential medium supplemented with 10% fetal bovine serum for 3 d. At harvest, cells were removed and washed with Hanks solution. An aliquot of cells was removed for protein determination, and the remainder was used for analysis of fatty acids after conversion to their methyl esters (17). Radiolabeled fatty acid methyl esters were separated by reversed-phase high performance liquid chromatography (18), and collected by a fraction collector. The radioactivity was counted by liquid scintillation counting. [Pg.284]
