Sarco-endoplasmic reticulum SERCA

Cyclopiazonic acid is a potent inhibitor of calcium uptake and acts as a selective inhibitor of the sarco-endoplasmic reticulum Ca2+ATPases (SERCAs) [53], it induces charge alteration in plasma membranes and mitochondria and it can function as an antioxidant [54].

The anthranilamide, DP-012, stimulates a rise in [Ce ]i under conditions of Ca -free saline similar to that observed with standard saline, indicating that this chemistry mobilizes calcium from internal calcium stores rather than through external entry via voltage-gated Ca channels. Three possible targets are involved in release of internal calcium stores, inositol trisphosphate receptors (IP3RS), ryanodine receptors (RyRs), and sarco-endoplasmic reticulum ATPase (SERCA). The endoplasmic reticulum takes up Ca " " using the sarco(endo)plasmic reticulum ATPase (SERCA) pump (Figure 11.4). The SERCA pump of many types of muscle is regulated by a protein called phospholambin, which binds to SERCA both in its cytosolic and its transmembrane regions, maintaining the pump in an inactivated state when in its nonphosphorylated form, but detaches from the pump upon phosphorylation, presumably due to a conformational change. In this sense, the SERCA pump is like the PMCA pump, except that Ca -saturated calmodulin activates PMCA, whereas kinase-dependent phosphorylation is involved in SERCA activation (perhaps also dependent on calmodulin).
