In this chapter, the different steps in the assessment of mixture exposure are discussed. The chapter starts from emission scenarios and subsequently discusses transformation processes taking place in the environment and their effects on mixture composition. Next, bioavailability is discussed, and exposure scenarios for both humans and biota in the environment are described. These descriptions also consider methods to assess exposure to mixtures. Most data available on mixture exposure are restricted to North America and Europe, but we recognize that there are emerging problems in other regions of the world. We restrict our discussion to man-made chemicals and those natural chemicals subject to regulation (metals, polycyclic aromatic hydrocarbons (PAHs)), because these represent the most well-studied group and the current priorities for risk assessment. [Pg.3]

Consequently the parameter used is the Specific Instantaneous Toxic Potential (SITP) of the material, which takes account of the specific nature of individual radionuclides in terms of their half life, persistence in the body etc, and calculates a theoretical dose which is a better measure of the potential harm to man. Costs and burdens of finding information, the nature of the PMN substance, anticipated manufacturing quantities, anticipated profits, and the size of the entity submitting the PMN are factors that can be considered in determining whether information is reasonably ascertainable. Because these are all subjective criteria, there is no definition of reasonably ascertainable that is appHcable in all situations. In fact the transition from bench to market of combination products is often hindered by a series of scientific and economic issues. Both the determination of the ideal cell type and the ideal biomaterial for the specific application can be problematic. Often, the use of autologous differentiated cells would be the best solution, but their use may not be feasible because of limitation on isolation and expansion. Moreover, the use of autologous cells also can introduce infections risks deriving from the use of xenogenic factors or animal feeder layers in culture. Hopefully, adult and embryonic stem cells may provide alternative solutions. Also the determination of the most suitable biomaterial for the specific application is a challenge. Most of the currently available synthetic materials are subjected to a foreign body reaction that can lead to serious complications when implanted in the human body. The use of natural scaffolds circumvents this problem, but it introduces other drawbacks such as inappropriate mechanical properties.
