The QA unit should have written procedures (SOPs) for the conduct of inspections and audits. These procedures should incorporate all considerations for the review of electronic data systems. The QA unit SOPs should address the role and responsibilities of the QA unit in software development, purchase, and validation activities, in-process audit procedures for data collected on line, procedures for on-line review of data (i.e., what will be verified and how much data will be reviewed), and the procedure for auditing reports using on-line data. 

In a review of data on occupational chemicals that may contaminate breast milk (Byczkowski et al. 1994), it is stated that lead may be excreted in milk in amounts lethal to the infant and that the metal may be mobilized from bone stores to milk during the lactation period. Even when the concentration of lead in mother s milk is low, the absorption of metals into the systemic circulation of infants is generally high when they are on a milk diet. To better understand the sensitivity of the nursing infant to chemicals, epidemiological studies, chemical monitoring, and model development and application are needed. 

The most reliable sources of data are the original citations of valuable experimental data in the reviewed scientific literature. Particularly reliable are those papers which contain a critical review of data from a number of sources as well as independent experimental determinations. A review of data from 250 chemicals found an 82% concordance between results of carcinogenicity testing in the mouse and the rat (Purchase, 1980). Haseman et al. (1984a) reported a concordance of 73% for 60 compounds studies in both species. However, 30 to 40% of 186 National Cancer Institute (NCI) chemicals were found to be positive in one species and negative in the other (Gold et al., 1984). It is reasonable to conclude that neither rodent species will always predict the results in the other rodent species or in humans, and that the use of two species will continue until we have a much better understanding of the mechanisms of carcinogenesis.

In the following sections of this article we shall deal with investigations concerning the questions enumerated above. In order to obtain an effective basis, it seems appropriate here to complement the review of data from older studies given in this introduction by a selection of some pertinent results and novel reactions from recent investigations. 

The use of microdialysis has enabled unbound drug concentrations to be determined in ECF, providing another measurement of penetration across the blood-brain barrier and one more closely related to activity. A review of data obtained by microdialysis [7], showed that free drug exposure in the brain is equal to or less than free drug concentration in plasma or blood, with ratios ranging from 4% for the most polar compound (atenolol) to unity for lipophilic compounds (e.g. carbamazepine). This largely supports the similar conclusions from the CSF data shown above. This relationship is illustrated in Figure 4.4.

They also reported that on the basis of a review of data on over 2000 chemicals that have been tested in various in vitro and/or in vivo assays, all chemicals showing estrogenic properties contained a ring structure. A protocol provides a mechanism for review of data and reports during, and at the conclusion of, the study. A regular review of the actual study conduct, 1n reference to the planned conduct as detailed 1n the protocol, provides a degree of confidence that the results obtained will be consistent with the study objectives. That 1s not to say that minor deviations can not, or should not, occur throughout the course of the study. The GLP regulations, however, require that these deviations be noted 1n the final report of the study. The study protocol provides the master" reference for compilation of these deviations.

The different models show bias. Concentration addition implies that effects always increase with each additional compound (even when present at extremely low concentrations), and there is clear evidence that this occurs with similarly acting compounds, but it is not yet clarified with dissimilarly acting compounds (see reviews, above). Thus, applying concentration addition to mixtures of dissimilarly acting compounds might overestimate effects and risks, and this may be an undesired feature for various risk assessments (e.g., for retrospective, diagnostic risk assessments).
