Regional assessments description

The introduction of 2AP in so many different structural contexts in the TPP riboswitch provides an opportunity to use it as a probe of the folding at both local and global levels. From the perspective of each local structure and dynamics, what is lacking is time-resolved data that would show how the 2AP is contained within the structure, and by implication how flexible that particular region of the RNA is in the free and bound states. The intrinsic flexibility of an RNA can be a critical part of its function, but is a parameter that is often difficult to assess. With the introduction of 2AP, at least part of that description could be obtained. 

In this chapter, the different steps in the assessment of mixture exposure are discussed. The chapter starts from emission scenarios and subsequently discusses transformation processes taking place in the environment and their effects on mixture composition. Next, bioavailability is discussed, and exposure scenarios for both humans and biota in the environment are described. These descriptions also consider methods to assess exposure to mixtures. Most data available on mixture exposure are restricted to North America and Europe, but we recognize that there are emerging problems in other regions of the world. We restrict our discussion to man-made chemicals and those natural chemicals subject to regulation (metals, polycyclic aromatic hydrocarbons (PAHs)), because these represent the most well-studied group and the current priorities for risk assessment. 

A parallel effort has been the assignment of cause or risk using a WoE approach. Menzie et al. (1996) provides one of the earliest and clearest descriptions of the WoE approach to assigning causality. The usefulness of WoE has been extensively discussed in a series of recent papers (Burton et al. 2002a, 2002b, Chapman et al. 2002, Forbes and Calow 2002). Clearly this approach has proven useful. However, it requires improvement in order to address the needs of regional risk assessment. 

The model system allows also a basin wide description of the transport of suspended particulate matter (SPM), which can be transported near the seabed within the so-called fluffy layer and is strongly affected by surface waves in coastal waters. Understanding the settling and resuspension of SPM in response to waves and currents is of great importance to assess the transports of material discharged by rivers from the coastal regions to the final deposit in the sediments of central basins. 

In the spectrophotometric method, color is assessed by means of a standard absorption spectrophotometer. In this case, a description of the hue, intensity and brightness of a non-turbid colored solution is possible in the visible region (400-700 mn) of the electromagnetic spectrum. Since pH will have a significant impact on the color of certain dyes, the pH of the dye solution employed must be measured and reported. Public awareness of neurotoxic chemicals comes from media descriptions of mass chemical disasters involving hundreds, if not thousands, of victims. Many mass chemical disasters fueled environmental movements, forced legislation of new laws, changed public perceptions of government and industry, and caused adverse health outcomes for many individuals.
