Another example is the application of the Jones-Marres silver method for rapid staining of fungi in the brain tissue of immunocompromised patients (Boon et al., 1998). This procedure can be carried out using the MicroMED BASIC microwave lab station (Milestone, s.r.l., 24010 Sorisole, Italy). The lab station has software for reliable control of power, time, and temperature using infrared temperature control for no-touch temperature determination. It also has a 360-degree rotation carousel (no hot spots) and produces printouts of the temperature and power levels used during various microwave steps. 

Microwave heat can also be applied for rapid staining of frozen sections. Frozen-section diagnosis plays an important role in the evaluation of the operability of the patient and in the examination of resection margins. The preparation for diagnosis, for example of signet-ring cell carcinoma in the peritoneum, can be accomplished in as brief a time as 30 sec by the modified periodic acid-Schiff s (PAS) reaction facilitated by microwave heating...

The initial steps of solubilization, DEAE chromatography and gel filtration were slight modifications of reported procedures [19]. The pooled fractions from a Sephacryl S-200 (Pharmacia) column were applied to a Mono Q HR5/5 FPLC column (Pharmacia) and eluted in a 20 ml gradient from 0-350 mM NaCl in 0.25 M sucrose-20 mM Tris-HCl pH 7.3. Fractions of 1 ml were collected and assayed for binding of NAA-1-[ C] (61 mCi/mmol, Amersham) by one of three methods [20]. The most active fractions were pooled, desalted and lyophilized. This preparation (approx. 50% receptor) was used either for monoclonal antibody production or was fully purified by native PAGE in a neutral pH discontinuous system [3]. The gel was briefly electroblotted (5 min, 10 mA) to nitrocellulose and the small fraction of transferred proteins visualized by rapid staining [8]. This blot was then used to locate precisely the bulk protein bands remaining in the gel. 

New sensitive assays for cellulase activity use as substrates modified carboxy-methylcelluloses containing either 2,4,6-trinitrophenyl groups (for spectrophoto-metric assay) or fluorescamine groups (for fluorometric assay). A simple and rapid staining procedure for qualitative and quantitative determinations of plant Citrus sinensis) and fungal T. viride) cellulases is based on the incorporation of carboxymethylcellulose into polyacrylamide gels. After electrophoresis of the crude extract, the gel is incubated in a phosphate buffer, acidified, and treated with iodide-iodine no colour develops in areas containing cellulase activity.
