Purines antiviral effect

Didanoside, a purine analog with antiviral effects (75 mg/kg p.o. q. 12 hours), is used in advanced human immunodeficiency virus (HIV) infection in patients who cannot tolerate or no longer respond to zidovudine therapy (see also Figure 107).

The condensation of various heterocycles (pyrimidine or purine) with allyl bromide has been effected in the presence of KI. This selectively afforded N-1 isomers with pyrimidines, but N-9, N-3 or N-9, N-7 isomer mixtures with purines (88MIP1 89MI12 91 Mil). Alkyl esters of 3-adenin-9-yl-2-hydroxypropanoic acids were reported as broad-spectrum antiviral agents (85JMC282). 

Inosine pranobex is a synthetic product, also known as isoprinosine or inosine dimepranol acedobene, with antiviral properties that are assumed to be related to its effect on T cell-mediated immunity rather than to direct antiviral activity. It has been tried in a wide range of viral diseases and also in rheumatoid arthritis (1), multiple sclerosis (2), and alopecia (3). However, clinical trials have mostly shown only modest therapeutic benefit or none at all (1,4), and no specific adverse effects, except for an increase in serum uric acid concentrations (5), reflecting the metabolic pathways of purines (6). 

Several interesting organoselenium compounds exhibit antiviral activities. One such is selenazofurin 113, effective against different types of viruses (herpes simplex, parainfluenza and rhinovirus).
