Although ribozymes have been shown to work in vitro in cellular assays, there are few reports demonstrating their efficacy in vivo. One such success utilized the rabbit model of IL-l-induced arthritis. In this model, IL-1 is believed to generate the formation of a proteinase mediator called stromelysin, which participates in the inflammatory condition. The intra-articular administration of ribozymes directed against stromelysin mRNA has been reported to produce a reduction in the message for this mediator in synovial fluid. [Pg.278]

In rheumatoid arthritis, collagenous tissues of the joint (including articular cartilage) are eroded by collagenases and neutral proteinases derived from proliferating cells of the synovial membrane and leukocytes. Therapeutic approaches may depend on regulation of collagen synthesis, breakdown, or both. [Pg.179]

Connective tissue destruction during inflammatory diseases, such as chronic wound, chronic leg ulcers, or rheumatoid arthritis, is the result of continuous supply of inflammatory cells and exacerbated production of inflammatory cytokines and matrix proteinases (Senni et al., 2006). According to the Senni et al. (2006), fucoidan from Ascophyllum nodosum is a potent modulator of connective tissue proteolysis. Further, the authors suggested that fucoidan could be used for treating some inflammatory pathology in which uncontrolled extracellular matrix degradation takes place. [Pg.173]

Hydroxamic acids, which have been extensively studied as metalloenzyme inhibitors and have been shown to be potent inhibitors of matrix metallo-proteinases, are a family of zinc-dependent enzymes associated with diseases such as cancer, arthritis and multiple sclerosis. [Pg.26]
