Bisphosphonates (particularly clodronate) have been shown to have anti-inflammatory effects in animal models of rheumatoid arthritis (RA), as well as in arthritis in humans. In adjuvant- and antigen-induced arthritis in rats, clodronate suppresses the inflammatory articular lesions in the inflamed joints [29], whilst in human RA, clodronate decreases the levels of interleukin (ILJ-1, tumor necrosis factor-alpha (TNFaand /1-microglobulin in the circulation [30]. In vitro, clodronate inhibits cytokine and nitric oxide (NO) release and inducible nitric oxide synthase (iNOS) expression in macrophage-like cells. [Pg.382]

As compared to control animals, significantly less arthritis was observed in mice with antigen-induced arthritis subcutaneously administered 2.5 mg/kg of an aqueous Iceland moss extract twice a week for 14 days before arthritis was induced and continuing for 36 days after (Freysdottir et al. 2008). [Pg.192]

Antigen-induced arthritis (AIA) is a model to induce arthritis in different species, mainly in mice, that was described in 1970s (Brackertz, MitcheU, Mackay, 1977 Cooke, Hurd, Ziff, Jasin, 1972), and it is stiU very useful to investigate mechanisms associated with joint inflammation. AIA may also be used as a proof-of-concept model for the development of new compounds in the preclinical phase. One of the most important features of joint inflammation in AIA is the high number of neutrophils that... [Pg.266]

The anti-inflammatory activity and therapeutic potential of evasins have been demonstrated in animal models [67]. Evasin-1 caused a significant reduction in skin inflammation in D6 mice sensitized with TPA and reduced lethahty, decreased leukocyte infiltration and preserved the lung architecture in a lung injury model. Evasin-3 was tested in a mouse model of antigen-induced arthritis and it was found to reduce leukocyte infiltration in the synovial cavity and inflammation. Both evasin-1 and evasin-3 reduced lethality in an intestinal ischemia model. [Pg.367]
