Fluorescence Spectroscopy.—Fluorescence studies can be used to obtain information about proteins in a number of ways. Firstly there is the use of fluorescent labels or probes in order to obtain quantitative analytical results. For example, Lockridge and Le Du used the fluorescent probe -methyl 7-(dimethyl-carbamoxy)quinolinium iodide to establish that there were four active sites in the tetrameric cholinesterase molecule from human serum. Weiel et labelled the initiation factor protein (IF3) with fluorescein to study its binding to the 30 S ribosomal subunits and demonstrated 1 1 stoicheiometry. [Pg.176]

Spectroscopic studies were performed on water in supercritical CO2 microemulsions using an ammonium carboxylate PFPE surfactant (24). FTIR spec-toscopy was used to identify a bulk water phase within the microemulsion capable of solubilizing ionic species and supporting inorganic reactions. In addition, the UV-visible spectrum of the solvatochromic probe methyl orange indicated three microenvironments within the microemulsions (a) a polar microenvironment like that found in dry PFPE reverse micelles (b) bulk water microenvironment and (c) an acidic microenvironment due to CO2 dissolved in water. [Pg.266]

The Diels-Alder reaction provides us with a tool to probe its local reaction environment in the form of its endo-exo product ratio. Actually, even a solvent polarity parameter has been based on endo-exo ratios of Diels-Alder reactions of methyl acrylate with cyclopentadiene (see also section 1.2.3). Analogously we have determined the endo-exo ratio of the reaction between 5.1c and 5.2 in surfactant solution and in a mimber of different organic and acpieous media. These ratios are obtained from the H-NMR of the product mixtures, as has been described in Chapter 2. The results are summarised in Table 5.3, and clearly point towards a water-like environment for the Diels-Alder reaction in the presence of micelles, which is in line with literature observations. [Pg.137]
